实时荧光定量PCR法用于流感病毒检测的效果分析 被引量:1

摘要 目的分析实时荧光定量聚合酶链式反应(PCR)法检测流行性感冒(流感)病毒的应用效果。方法250例疑似流感患者,取患者咽拭子标本,分别应用病毒分离培养法和实时荧光定量PCR法进行检测。对比两种检测方法的阳性检出率、灵敏度、特异度及准确率。结果实时荧光定量PCR法阳性检出率37.20%高于病毒分离培养法的23.60%,差异有统计学意义(P<0.05)。实时荧光定量PCR法的灵敏度62.14%、特异度94.55%及准确率76.40%均高于病毒分离培养法的31.43%、86.36%、55.60%,差异有统计学意义(P<0.05)。结论相较于病毒分离培养法,通过实时荧光定量PCR法检测流感病毒能更快更准确检测样本,具有较高的应用价值。 Objective To analyze the effect of real-time fluorescent quantitative polymerase chain reaction(PCR)in the detection of influenza virus.Methods In 250 patients with suspected influenza,throat swab samples were taken from patients and tested by viral isolation and culture method and real-time fluorescence quantitative PCR method,respectively.The positive detection rate,sensitivity,specificity and accuracy of the two assays were compared.Results The positive detection rate of real-time fluorescent quantitative PCR method was 37.20%,which was higher than 23.60%of virus isolation and culture method,and the difference was statistically significant(P<0.05).The sensitivity 62.14%,specificity 94.55%and accuracy 76.40%of real-time fluorescence quantitative PCR method were higher than 31.43%,86.36%and 55.60%of viral isolation and culture method,and the difference was statistically significant(P<0.05).Conclusion Compared with viral isolation and culture method,the real-time fluorescence quantitative PCR can detect influenza virus more quickly and accurately,which has high clinical application value.
