分光光度法测定破壁灵芝孢子粉中三萜类化合物的含量 被引量:1

摘要 目的:建立破壁灵芝孢子粉中三萜及甾醇含量的测定方法。方法:以齐墩果酸为对照品,用5%香草醛冰醋酸、高氯酸显色,在546 nm波长处测定样品吸光度。结果:齐墩果酸在0.01834~0.0917 mg·mL^(-1)线性关系良好,回归方程为y=7.54560x-0.01161,相关系数R^(2)=0.99954,回收率为95.52%~101.29%,均值为98.55%,RSD为2.5%,方法重复性试验测定样品含量值范围为3.66%~3.85%,均值为3.76%,RSD为1.9%。结论:该方法操作简便、快速,结果稳定,重现性好,适用于破壁灵芝孢子粉中三萜及甾醇含量的测定。 Objective:To establish a method for the determination of triterpenoids and sterols in Sporoderm-broken Ganoderma lucidum spores powder.Methods:Using oleanolic acid as reference,5%vanillin glacial acetic acid and perchloric acid were used to color reaction.The absorbance of the sample was measured at 546 nm.Results:The linear range of oleanolic acid was 0.01834~0.0917 mg·mL^(-1),the regression equation was y=7.54560x-0.01161,the correlation coefficient R^(2)=0.99954,the recovery rate was 95.52%~101.29%,the mean value was 98.55%,the RSD was 2.5%,and the linear range was 0.01834~0.09170 mg·mL^(-1).The method repeatability test ranged from 3.66%~3.85%,with an average of 3.76%and a RSD of 1.9%.Conclusion:The method is simple,rapid,stable and reproducible.It is suitable for the determination of triterpenoids and sterols in Sporoderm-broken Ganoderma lucidum spores powder.
