假酸浆毛状根黄酮提取工艺优化及其抑菌活性研究

摘要 分析了假酸浆毛状根中黄酮类化合物的超声辅助提取技术,对抑菌活性物质的提取与测定进行分析。优选出黄酮的最佳提取条件为:乙醇浓度为70%,料液配制比为1∶20,提取时间控制在40 min,提取温度设置在70℃,得到黄酮提取率为8.3%。采用滤纸片法对黄酮进行抑菌活性检测,测得提取出的黄酮对大肠杆菌抑菌浓度最低为1 mg/mL,对金黄色葡萄球菌抑菌浓度最低为1.5 mg/mL,且随异黄酮浓度升高抑菌作用越明显,说明假酸浆毛状根中黄酮是一种具有抑菌活性的物质,可作为一种天然有机防腐剂。 The research analyzes the assistance extraction technology of ultrasonic wave on flavonoid from Nicandra hairy root,and the extraction and measurement of bacteriostatic activity.The optimal extraction condition of Nicandra is as following:the ethanol concentration is 70%,the ratio of liquid to material is 1∶20,the extraction time is controlled within 40min,and the extraction temperature is set at 70℃,and Nicandra extraction rate is 8.3%.Filtering paper method is used to test the bacteriostatic activity on flavone,the lowest inhibitory concentration of escherichia coli on flavone is 1 mg/mL,and the lowest inhibitory concentration to Staphylococcus aureus is 1.5mg/mL.As the concentration of isoflavone increases,the antibacterial effect becomes more obvious.This indicates that flavone in Nicandra hairy root flavonoid is a kind of baceriostatic activity material,and it can be regarded as a kind of natural organic antiseptic substance.
