石柱参SzPLC1基因的生物信息学分析及胁迫应答

摘要 利用RT-PCR结合RACE技术,克隆石柱参磷脂酶C(SzPLC1)基因,并采用生物信息学方法对该基因及其编码蛋白进行数据分析,同时探讨干旱胁迫对SzPLC1表达的影响。结果显示:该基因全长1725 bp,开放阅读框(ORF)长度为1455 bp,编码484个氨基酸,分子质量为54.8 ku,具有2个跨膜结构域X和Y区;SzPLC1蛋白二级结构包括无规则卷曲(40.91%)、α-螺旋(37.54%)、延伸链(18.39%)和β-转角(4.96%)。系统进化分析表明石柱参SzPLC1蛋白与萝卜(Raphanus sativus,XP_018480211.1)的RsPLC1蛋白表现出较高的同源性(95%)。String蛋白互作网络分析表明,与SzPLC1蛋白互作的蛋白质共有10个,均为磷脂酰肌醇信号通路的重要酶类,说明SzPLC1是磷脂酰肌醇信号通路中关键的酶。qRT-PCR分析表明,在干旱处理下,SzPLC1的表达量明显上调,说明SzPLC1蛋白具有抗干旱胁迫的功能。 Phospholipase C(SzPLC1)gene of Shizhu ginseng was cloned by RT-PCR combined with RACE technology,and bioinformatics methods were used to analyze the gene and its encoded protein.Meanwhile,the effect of drought stress on the expression of SzPLC1 was discussed.The results showed that full-length of SzPLC1 gene was 1725 bp,and the ORF was 1455 bp,which encodes 484 amino acids with a molecular weight of 54.8 ku.The gene had two transmembrane domains,namely X and Y region.The secondary structure of SzPLC1 protein included random coil(40.91%),α-helix(37.54%),extended strand(18.39%)andβ-turn(4.96%).The phylogenetic analysis showed that SzPLC1protein had high homology(95%)with RsPLC1 protein of radish(Raphanus sativus,XP_018480211.1).The analysis of string protein interaction network showed that there were 10 proteins interacting with SzPLC1 protein,which were all important enzymes in phosphatidylinositol signaling pathway,indicating that SzPLC1 was the key enzyme in phosphatidylinositol signaling pathway.qRT-PCR analysis showed that the expression of SzPLC1 was significantly up-regulated under drought stress,indicating that SzPLC1 protein has the function of drought stress.
