维药土提汤水提物和醇提物抗炎镇痛作用及其对急性咽炎的治疗作用比较

Comparison of anti-inflammatory and analgesic effects of water extract and alcohol extract of Uyghur Tuti Decoction and their therapeutic effects on acute pharyngitis

[目的]研究土提汤水提物和醇提物的抗炎镇痛作用,并比较两者对急性咽炎的治疗作用,为其药用开发奠定基础。[方法]采用小鼠热板实验、小鼠甩尾实验、二甲苯致小鼠耳肿胀实验、小鼠腹腔血管通透性实验和完全弗氏佐剂致大鼠足肿胀实验分析抗炎镇痛效果,同时采用大鼠急性咽炎实验考察土提汤水提物和醇提物对急性咽炎的治疗效果。[结果]与模型组比较,阳性对照组、土提汤水提取物组和醇提取物组可显著提高小鼠痛阈值,降低小鼠腹腔血管通透性,降低二甲苯所致的耳肿胀,降低完全弗氏佐剂所致的足肿胀,差异具有统计学意义(P<0.05);水提物与醇提物相比,醇提物的抗炎镇痛效果更好。土提汤各剂量组均能明显减轻咽黏膜上皮的角化和钉突,减少炎细胞浸润;缩小肺组织的肺泡间隙,减轻肺泡扩大现象,减少肺泡腔内炎细胞浸润。与模型组比较,土提汤水提物及醇提物高低剂量组肿瘤坏死因子-α(TNF-α)含量显著降低(P<0.01)。[结论]土提汤提取物对大鼠急性咽炎模型具有良好的治疗效果,且对急性咽炎的治疗作用是通过减少肿胀、渗出和镇痛来实现的。土提汤醇提物镇痛消炎作用强于水提物,即醇提法为土提汤镇痛消炎作用的最佳提取方式,为后续剂型改进提供实验依据。

[Objective]To study the anti-inflammatory and analgesic effects of Tuti decoction water extract and Tuti Decoction alcohol extract,and their therapeutic effects on acute pharyngitis were compared to lay experimental foundation for their medicinal development.[Methods]The anti-inflammatory animal models were used to figure out the effects of different extracts of Tuti Decoction on acute inflammation.Hot-plate procedure and writhing test in mice were adopted in experimental analgesic models,anti-inflammatory model of mouse ear edema induced by xylene,rat paw edema induced by complete Freund’s adjuvant and exudation of abdominal blood capillaries of mice were applied.Acute pharyngitis model in rats were adopted to investigate the treatment effect of Tuti Decoction water extract and Tuti Decoction alcohol extract.[Results]Compared with the model group,the positive group,the Tuti Decoction water extract group and Tuti Decoction alcohol extract group could significantly improve the pain threshold,reduce the abdominal vascular permeability of mice,reduce the ear swelling,and reduce the foot swelling(P<0.05).The anti-inflammatory and analgesic effect of alcohol extract is better than that of water extract.Tuti Decoction can obviously reduce the keratinization of pharyngeal mucosa epithelium,reduce the infiltration of inflammatory cells,reduce the alveolar space of lung tissue,and reduce the infiltration of inflammatory cells in alveolar cavity.Compared with the model group,the content of TNF-αin the high and low dose group was significantly decreased(P<0.01).[Conclusion]Tuti Decoction extract has a good therapeutic effect on acute pharyngitis in rats,and the therapeutic action on acute pharyngitis is achieved by reducing swelling,reducing exudation and relieving pain.The analgesic and anti-inflammatory effect of alcohol extract of Tuti Decoction is stronger than water extract,which lays experimental foundation for the subsequent formulation improvement.
