异土木香内酯对人舌鳞癌Tca8113细胞的增殖、迁移和侵袭的影响

The Effect of Isoalantolactone on Proliferation,Migration and Invasion of Human Tongue Squamous Cell Carcinoma Tca8113 Cells

目的研究异土木香内酯对舌鳞癌细胞Tca8113的增殖、迁移和侵袭能力的影响。方法用CCK-8法检测异土木香内酯作用于舌鳞癌Tca8113细胞的半数抑制浓度(IC50),EDU实验方法检测异土木香内酯抑制Tca8113细胞增殖能力。检测土木香内酯对Tca8113细胞凋亡影响实验。Western Blot检测异土木香内酯对Tca8113细胞凋亡蛋白表达的影响。结果实验结果表明,异土木香内酯作用于舌鳞癌Tca8113细胞的半数抑制浓度(IC50)为20μM,20μM异土木香内酯可以抑制舌鳞癌Tca8113细胞活性(P<0.01),细胞增殖能力(P<0.01);进一步结果表明,异土木香内酯还可以抑制对Tca8113 P迁移,同时降低Tca8113细胞侵袭能力(P<0.01);促进Tca8113细胞凋亡的影响(P<0.01)。Western Blot实验结果表明,异土木香内酯可以下调舌癌Tca8113细胞的Bcl-2蛋白,上调caspase3蛋白的表达。结论实验结果表明异土木香内酯对舌癌Tca8113细胞有抑制作用。

Objective To study the effect of isoalantolactone on the proliferation,migration and invasion of tongue squamous cell carcinoma Tca8113 cells.Methods CCK-8 method was used to detect half of inhibitory concentration(IC50)of isoalantolactone on Tca8113 cells,and EDU experiment was used to detect the inhibitory effect of isoalantolactone on the proliferation of Tca8113 cells.The effect of isoalantolactone on apoptosis of Tca8113 cells was tested.The effect of isoalantolactone on the expression of apoptosis protein in Tca8113 cells was detected by Western Blot.Results The experimental results showed that half of inhibitory concentration(IC50)of isoginolactone on tongue squamous cell carcinoma Tca8113 cells was 20μM,which could inhibit the activity and cell proliferation of tongue squamous cell carcinoma Tca8113 cells(P<0.01).Further results showed that isoginolactone could inhibit the migration of Tca8113 cells,decrease the invasive ability of Tca8113 cells and promote Tca8113 cell apoptosis(P<0.01).The results of Western Blot experiments show that isoalantolactone can downregulate Bcl-2 protein and upregulate caspase3 protein expression in tongue cancer Tca8113 cells.Conclusion The experimental results show that isoalantolactone has an inhibitory effect on tongue cancer Tca8113 cells.
