银香解毒膏通过抑制炎症反应减轻皮肤金黄色葡萄球菌感染和促进毛囊生长

Yinxiang Jiedu ointment alleviates skin staphylococcus aureus infection and promotes hair follicle growth by inhibiting inflammation

目的探究银香解毒膏抑制皮肤金黄色葡萄球菌感染、促进皮肤毛囊修复的机制。方法使用"胶带剥脱法"建立小鼠金黄色葡萄球菌感染模型,分别在涂抹药银香解毒膏、莫匹罗星膏、赋形剂膏治疗后的第3d和第5d皮肤取样。免疫荧光染色观察中性粒细胞招募,Western blot检测NF-κB(p-p65)和BAX/BCL2水平,EdU染色示踪毛囊细胞增殖。结果相比于莫匹罗星膏,银香解毒膏能更好地治疗被金黄色葡萄球菌感染的皮损,促进皮肤损伤部位快速愈合,减低皮损部位中性粒细胞的招募,抑制NF-κB以及凋亡信号通路激活,促进感染部位皮肤毛囊细胞的增殖。结论银香解毒膏可以降低金黄色葡萄球菌对皮肤的损伤程度,对皮肤深层的毛囊组织具有保护作用,促进毛囊的新生。

Objective To investigate the mechanism of Yinxing Jiedu ointment in inhibiting skin Staphylococcus aureus infection and promoting skin hair follicle repair.Methods The “tape stripping method” was used to establish a mouse model of Staphylococcus aureus infection.Yinxiang Jiedu ointment,mupirocin ointment,and excipient ointment were applied on the skin lesions of the corresponding group.Skin samples were obtained on the third day and the fifth day.Immunofluorescence staining was used to observe the recruitment of neutrophils,Western blot was used to detect the activation of NF-κB(p-p65) and the apoptosis signaling pathway(BAX/BCL2),and EdU staining was used to observe the proliferation of hair follicle cells.Results Compared with mupirocin ointment,Yinxiang Jiedu ointment can more effectively protect the skin lesions infected by Staphylococcus aureus,promote the rapid healing of skin lesions,reduce the recruitment of neutrophils in the lesions,inhibit the activation of NF-κB and the apoptosis signaling pathway,and promote the proliferation of skin hair follicle cells at the infected site.Conclusion Yinxiang Jiedu ointment can alleviate skin damage caused by Staphylococcus aureus,protect the deep hair follicle tissue,and promote the regeneration of hair follicles.
