盐胁迫下拟南芥去乙酰化酶基因的转录组分析 被引量:1

摘要 为了探究盐胁迫下拟南芥HD2去乙酰化酶调控的下游基因,本研究以拟南芥野生型WT和四突(hd2q,HD2四个基因的突变体)为材料,采用RNA-seq技术和生物信息学方法,对生长10 d的幼苗在高盐(150 mmol/L NaCl)处理前和后进行比较转录组分析,并结合实时荧光定量PCR验证转录组数据的可靠性.结果显示:在高盐处理前WT和hd2q共有25个差异基因,其中上调基因2个,下调基因23个.在高盐处理后,WT和hd2q共有1407个差异基因,其中上调基因772个,下调基因635个.GO和KEGG分析显示,对照组的差异基因主要富集在胞外区域和响应脱落酸上,盐处理组的差异基因主要富集在胞外区域和细胞壁上,并且发现有41个与盐响应途径相关的差异基因(27个上调基因,14个下调基因).结果表明拟南芥HD2去乙酰化酶调控下游盐响应基因从而使拟南芥适应盐胁迫环境. In order to explore the downstream genes regulated by HD2 deacetylase in Arabidopsis thaliana under salt stress,wild-type and hd2q(HD2 four mutants)of Arabidopsis thaliana were used in this study.RNA-seq and bioinformatics methods were used to analyze the transcriptome of 10 days old seedlings before and after high salt(150 mmol/L NaCl)treatment,and the reliability of transcriptome data was testified by real-time quantitative PCR.The results showed that there were 25 differentially expressed genes in WT and hd2q,including 2 up-regulated genes and 23 down-regulated genes.After high salt treatment,there were 1407 differentially expressed genes in WT and hd2q,including 772 up-regulated genes and 635 down-regulated genes.GO and KEGG analysis showed that the differential expressed genes in the control group were mainly enriched in the extracellular region and response to abscisic acid,while the differential expressed genes in the salt treatment group were mainly enriched in the extracellular region and cell wall,and 41 differentially expressed genes(27 up-regulated and 14 down-regulated)in salt-responsive pathways were identified.Our results suggest that HD2 deacetylase regulates downstream salt responsive genes to adapt Arabidopsis to salt stress.
