乙型病毒性肝炎患者血清微小RNA-101、脂肪酸结合蛋白4 mRNA、脂肪酸结合蛋白5 mRNA表达水平与肝纤维化程度的相关性 被引量:2

摘要 目的探讨乙型病毒性肝炎患者血清微小RNA-101(miR-101)、脂肪酸结合蛋白4(FABP4)mRNA、脂肪酸结合蛋白5(FABP5)mRNA水平与肝纤维化程度的相关性。方法选取2020年3月至2021年3月于河南大学第一附属医院确诊的96例乙型病毒性肝炎患者,纳入乙型肝炎组。选取同期该院健康体检者94例作为对照组。根据乙型病毒性肝炎患者肝纤维化程度将其分为S 0期(40例)、S_(1)期(28例)、S_(2)期(16例)、S_(3)期(12例)。采用实时荧光定量聚合酶链式反应(qRT-PCR)法检测血清miR-101、FABP4 mRNA、FABP5 mRNA水平。采用Pearson法分析乙型病毒性肝炎患者血清miR-101与FABP4 mRNA、FABP5 mRNA水平的相关性。采用Spearman法分析血清miR-101、FABP4 mRNA、FABP5 mRNA水平与乙型病毒性肝炎患者肝纤维化程度的相关性。结果乙型肝炎组血清FABP4 mRNA、FABP5 mRNA水平高于对照组,miR-101水平低于对照组(P<0.05)。血清FABP4 mRNA、FABP5 mRNA水平随着肝纤维化分期的增加而上升,miR-101水平随着肝纤维化分期的增加而下降(P<0.05)。乙型病毒性肝炎患者血清miR-101与FABP4 mRNA、FABP5 mRNA水平均呈负相关(r=-0.524、-0.590,P<0.05)。乙型病毒性肝炎患者肝纤维化程度与血清FABP4 mRNA、FABP5 mRNA均呈正相关(r=0.654、0.723,P<0.05),与miR-101呈负相关(r=-0.861,P<0.05)。结论血清miR-101、FABP4 mRNA、FABP5 mRNA水平与乙型病毒性肝炎患者肝纤维化程度存在相关性,可能对评估肝纤维化程度有潜在价值。 Objective To investigate the correlation between the levels of serum microRNA-101(miR-101),fatty acid-binding protein 4(FABP4)mRNA and fatty acid-binding protein 5(FABP5)mRNA and the degree of liver fibrosis in patients with viral hepatitis B.Methods Ninety-six patients with viral hepatitis B diagnosed in the First Affiliated Hospital of Henan University from March 2020 to March 2021 were selected and included in hepatitis B group.During the same period,94 healthy examined people in this hospital were selected as control group.According to the degree of liver fibrosis,the patients with viral hepatitis B were divided into S 0 stage(40 cases),S_(1) stage(28 cases),S_(2) stage(16 cases)and S_(3) stage(12 cases).The levels of serum miR-101,FABP4 mRNA and FABP5 mRNA were detected by real-time quantitative polymerase chain reaction(qRT-PCR).Pearson method was used to analyze the correlation between serum miR-101 and the levels of FABP4 mRNA and FABP5 mRNA in patients with viral hepatitis B.Spearman method was used to analyze the correlation between the levels of serum miR-101,FABP4 mRNA and FABP5 mRNA and the degree of liver fibrosis in patients with viral hepatitis B.Results The levels of serum FABP4 mRNA and FABP5 mRNA in hepatitis B group were higher than those in control group,and the level of miR-101 in hepatitis B group was lower than that in control group(P<0.05).The levels of serum FABP4 mRNA and FABP5 mRNA increased with the increase of liver fibrosis stages,and the level of miR-101 decreased with the increase of liver fibrosis stages(P<0.05).The level of serum miR-101 was negatively correlated with FABP4 mRNA and FABP5 mRNA in patients with viral hepatitis B(r=-0.524,-0.590,P<0.05).The degree of liver fibrosis in patients with viral hepatitis B was positively correlated with serum FABP4 mRNA and FABP5 mRNA(r=0.654,0.723,P<0.05),and negatively correlated with miR-101(r=-0.861,P<0.05).Conclusion The levels of serum miR-101,FABP4 mRNA and FABP5 mRNA are correlated with the degree of liver fibrosis in patients with viral hepatitis B,and may be of potential value in evaluating the degree of liver fibrosis.
