蜂毒穴位注射对急性偏头痛大鼠行为学以及血浆降钙素基因相关肽、P物质含量的影响 被引量:2

Effect of Acupoint Injection of Bee Venom on Behavior and Plasma SP and CGRP Levels in Rats with Acute Migraine

摘要 目的观察蜂毒穴位注射对急性偏头痛大鼠行为学以及血浆降钙素基因相关肽(Calcitonin gene related peptide,CGRP)、P物质(SP)含量的影响,探讨蜂毒对偏头痛的作用机制。方法将健康雄性SD大鼠48只,随机分为空白组、模型组、佐米曲普坦组、蜂毒低、中、高3个剂量组,每组8只,后5组大鼠通过从后颈部皮下注射硝酸甘油10 mg/kg制作急性偏头痛大鼠模型,造模成功5 min后,给予相应药物干预,给药后2 h、4 h2个时间点取材,采用酶联免疫吸附测定法(Enzyme-Linked Immuno Sorbent Assay,ELISA)检测血浆SP、CGRP含量;并观察各组大鼠行为学表现。结果与空白组大鼠比较,模型组大鼠在0~30 min、30~60 min、60~90 min、90~120 min 4个时间段挠头次数、爬笼次数明显增多(P<0.01);与模型组比较,佐米曲普坦组、蜂毒中、高剂量组在0~30 min、30~60 min、60~90 min、90~120 min 4个时间段大鼠挠头次数、爬笼次数均明显减少(P<0.05,P<0.01);蜂毒低剂量组在0~30 min、30~60 min、60~90 min、90~120 min 4个时间段大鼠挠头次数明显减少,在0~30 min这个时间段大鼠爬笼次数明显减少(P<0.05,P<0.01);与空白组大鼠比较,模型组血浆SP、CGRP含量明显升高,说明偏头痛大鼠造模成功。与模型组比较,在2 h和4 h 2个时间点佐米曲普坦组、蜂毒中、高剂量组血浆中SP、CGRP含量均明显降低(P<0.05,P<0.01),蜂毒低剂量组血浆中CGRP含量明显降低(P<0.01);蜂毒中、高剂量组4 h比2 h血浆中SP、CGRP明显减少(P<0.05,P<0.01),蜂毒低剂量组4 h比2 h血浆中CGRP明显减少(P<0.05,P<0.01)。结论蜂毒穴位注射可明显改善大鼠行为学表现,其作用机制可能与减少大鼠血浆内SP、CGRP含量有关。 Objective To observe the effects of acupoint injection of bee venom on the behavior and plasma calcitonin gene related peptide(CGRP)and substance P(SP)levels in acute migraine rats,and to explore the effect of bee venom on migraine mechanism.Methods Forty-eight healthy male SD rats were randomly divided into three dose groups:blank group,model group,zolmitriptan group,bee venom low,medium,and high dose groups,with 8 in each group.A rat model of acute migraine was made by subcutaneous injection of nitroglycerin 10 mg/kg in the back neck.5 minutes after the model was successfully established,corresponding drug intervention was given.The samples were taken at 2 h and 4 h after administration,and the plasma SP and CGRP contents were detected by Enzyme-Linked ImmunoSorbent Assay(ELISA).The behavioral performance of rats in each group were observed.Results Compared with rats in the blank group,the rats in the model group had a significant increase in the number of head scratching and cage climbing in the four time periods of 0~30 min,30~60 min,60~90 min,and 90~120 min(P<0.01).Compared with the model group Compared with the four time periods of 0~30 min,30~60 min,60~90 min,and 90~120 min,the number of head scratching and the number of cage climbing in the zolmitriptan group,bee venom medium and high dose group were significantly reduced(P<0.05,P<0.01).In the low-dose bee venom group,the number of scratching heads in the four time periods of 0~30 min、30~60 min、60~90 min and 90~120 min was significantly reduced,and the number of climbing cages was significantly reduced during the time period of 0~30 min(P<0.05,P<0.01).Compared with the blank group,the plasma SP and CGRP levels in the model group were significantly increased,indicating that the migraine rats were successfully modeled.Compared with the model group,the levels of SP and CGRP in the plasma of the zolmitriptan group,medium and high-dose groups of bee venom were significantly reduced at 2 h and 4 h(P<0.05,P<0.01),and bee venom was low.The plasma levels of CGRP in the dose group were significantly reduced(P<0.01).The plasma SP and CGRP in the medium and high-dose groups of bee venom were significantly lower than that of 2 h in 4 h(P<0.05,P<0.01).The low-dose group of bee venom in 4 h compared with 2 h plasma CGRP decreased significantly(P<0.05,P<0.01).Conclusion Acupoint injection of bee venom can significantly improve the behavioral performance of rats,and its mechanism of action may be related to the reduction of SP and CGRP levels in rat plasma.
