TSWV侵染辣椒的胼胝质沉积及细胞病理学研究

摘要 【目的】研究番茄斑萎病毒(Tomato spotted wilt orthotospovirus,TSWV)侵染寄主植物辣椒(Capsicum annuum)时辣椒的防御反应,揭示其病变规律。【方法】摩擦接种TSWV,于接种后1、3、7、14和21 d采集辣椒系统叶片,利用苯胺蓝染色法和透射电子显微镜技术观察胼胝质沉积及超微结构变化。【结果】TSWV接种于辣椒后,辣椒细胞内观察到TSWV病毒粒子,在辣椒系统叶片气孔、筛管和细胞壁上均观察到胼胝质沉积,同时细胞超微结构发生病变,叶绿体结构被破坏,出现变形和解体现象,严重时引发细胞死亡。【结论】TSWV侵染辣椒后,辣椒启动胼胝质沉积进行防御反应,以阻止病毒进一步入侵;但同时也会引起细胞发生病变,导致植物出现褪绿、畸形和坏死等现象。 [Purpose]To study the defense response of Tomato spotted wilt orthotospovirus(TSWV)to Capsicum annuum,and to reveal the regularity of pathological changes.[Methods]The systemic leaves of C.annuum were collected at 1,3,7,14 and 21 days after friction inoculation by TSWV.The deposition of callose and the ultrastructural changes were observed by aniline blue staining and transmission electron microscopy.[Results]After inoculated TSWV on C.annuum,TSWV virus particles were observed in the cells,and callose deposition was observed in stomata,sieve tube and cell wall of C.annuum systemic leaves.Meanwhile,the ultrastructure of the cells was damaged,chloroplast structure was damaged,deformation and disintegration occurred,and the cell died seriously.[Conclusion]After TSWV infection,C.annuum initiated callose deposition to prevent further virus invasion;but at the same time,it can also cause cell lesions,leading to plant chlorosis,deformity and necrosis.
