参薯GRAS基因家族的挖掘与特征分析 被引量:1

Excavation and Characterization Analysis of GRAS Gene Family in Greater Yam(Dioscorea alata L.)

摘要 参薯属于薯蓣科薯蓣属,是热带地区重要的粮食作物,具有很高的营养价值。GRAS基因是一类重要转录因子,在植物的生长和发育中起关键作用。本研究从参薯基因组鉴定75个GRAS基因,通过多序列比对结果和系统进化树构建,将参薯GRAS家族蛋白划分为7亚家族,其中属于LISCL亚家族基因最多。分析Da GRAS保守motif发现,同一亚家族的GRAS蛋白具有更相似的motif结构、特征。根据参薯叶、块茎、零余子和花序组织转录谱数据分析GRAS基因的表达量,发现大比例基因表达量低,Da GRAS18、Da GRAS53等基因表达具有组织特异性。RT-qPCR实验表明DaGRAS9基因在块茎中高表达,其结果与转录谱数据一致,该基因的转录自激活实验表明该基因有转录活性。本研究对参薯Da GRAS家族进行初步分析,为以后Da GRAS家族的基因功能验证打下基础。 Greater yam(Dioscorea alata)belongs to the genus Dioscorea of the family Dioscoreaceae and is an important food crop in tropical and subtropical areas,which has high nutritional value.GRAS transcription factors play a key role in plant growth and development.The study used the genome of greater yam to identify 75 redundant sequences with GRAS domains.Based on the results of multiple sequence alignment and phylogenetic tree construction,the GRAS members were divided into seven subgroups,and most of them belonged to LISCL subfamily.The identification of conserved motif in GRAS protein demonstrated that the same subfamily of GRAS proteins had more similar motif structure.According to the transcriptomic datasets of leaf,tuber,and inflorescence tissues,it was found that the expression patterns of DaGRAS18 and DaGRAS53 were tissue-specific.RT-qPCR showed that DaGRAS9 was highly expressed in tuber,which was consistent with the transcriptional data.Auto-activation test inferred that DaGRAS9 had transcriptional activation ability.The study,identified and characterized GRAS transcription factor family in greater yam,which provided the basis for the further functional validation of Da GRAS.
