蒙药苏格木勒-3汤对IDCM心律失常作用研究 被引量:2

摘要 目的:探求蒙药苏格木勒-3汤(SD-3)对免疫性扩张型心肌病(IDCM)小鼠心律失常干预作用及其机制.方法:将30只小鼠随机分为对照组、IDCM组、蒙药SD-3组.其中IDCM组和蒙药SD-3组小鼠腹腔内注射0.1 mL10-5 TCID50CVB3,造模3个月后,蒙药SD-3组小鼠每天灌胃给予蒙药SD-3(15 mg·kg^(-1)),一天一次,而对照组及IDCM组则灌胃生理盐水,一天一次,灌胃4周.4周后,行心电图检测后高浓度CO2处死动物,4%多聚合甲醛(500 mL)经腔静脉灌流心脏,然后将心脏组织切片后HE、masson染色.采用免疫印迹的技术分析心肌组织中connexin43和connexin 40蛋白的表达情况.结果:与对照组相比,IDCM组小鼠出现明显心律失常;与IDCM小鼠相比,灌胃给予蒙药SD-3后,小鼠心律失常明显减少.与对照组相比,IDCM组小鼠心肌细胞缝隙连接蛋白CX40及CX43表达明显减少;与IDCM组小鼠相比,蒙药SD-3组小鼠心肌细胞缝隙连接蛋白CX40及CX43表达增加(P<0.01).结论:蒙药苏格木勒-3汤可以明显减少IDCM小鼠心律失常,其作用可能是通过增加缝隙连接蛋白CX40及CX43表达实现的. Objective:To explore the effect of Mongolian medicine Sugemule-3 decoction(SD-3)on arrhythmia mice with immune dilated cardiomyopathy(IDCM)and its mechanism.Methods:Thirty mice were randomly divided into a control group,an IDCM group and a Mongolian medicine Sugemule-3 decoction group,ten mice in each group.The IDCM group and Mongolian medicine SD-3 group were intraperitoneally injected with 0.1 mL10-5 TCID50CVB3.Three months after modeling,the Mongolian medicine SD-3 group was given Mongolian medicine SD-3(15 mg·kg^(-1))by gavage once a day,while the control group and IDCM group were given normal saline once a day,totally for four weeks.After four weeks,the mice were killed with high concentration CO2 after ECG detection.The whole heart was obtained by perfusing the heart with 4%paraformaldehyde(500 mL)through vena cava,then the heart tissue was sectioned and stained with HE and Masson.The expression of connexin(Cx)43 and connexin 40 in myocardium was analyzed by Western blot.Results:Compared with the control group,mice in the IDCM group had obvious arrhythmia.Compared with the IDCM group,the mouse arrhythmia was significantly reduced after intragastric administration of Mongolian medicine SD-3.Compared with the control group,the expression of connexin Cx40 and Cx43 in mouse cardiomyocytes in the IDCM group decreased significantly.Compared with the IDCM group,the expression of Cx40 and Cx43 in mouse cardiomyocytes in Mongolian medicine SD-3 group increased(P<0.01).Conclusion:Mongolian medicine Sugemule3 decoction can significantly reduce arrhythmia in mice with IDCM,which possibly be achieved by increasing the expression of Cx40 and Cx43.
