基于网络药理学探究补髓丹治疗骨质疏松的作用机制

摘要 目的基于网络药理学预测补髓丹治疗骨质疏松的主要活性成分及其作用的靶点,探讨其抗骨质疏松的作用机制。方法借助TCMSP数据库检索补髓丹的化学成分和作用靶点,通过Drug Bank、Gene Cards、Disgenet、TTD、OMIM等多个数据库查询与骨质疏松相关的基因。运用Cytoscape 3.2.1构建化合物-靶点网络、蛋白相互作用(PPI)网络筛选出核心靶点,最后通过MetaScape进行GO功能富集分析和KEGG通路富集分析,研究其作用机制。结果从补髓丹中筛选出了78个化合物307个相应蛋白靶点,PPI网络筛选出了包括MAPK1、TP53、ESR1和AKT1等在内的166个核心靶点,主要涉及了癌症信号通路、MAPK信号通路、凋亡信号通路、PI3K-Akt信号通路等。结论补髓丹是多成分、多靶点、整体调节来治疗骨质疏松的,通过网络药理学探讨了补髓丹抗骨质疏松的潜在机制,为其活性成分研究和进一步临床试验研究提供理论依据。 Objective Based on network pharmacology,to predict the main active components and targets of Busui mini-pills in the treatment of osteoporosis,and to explore its anti-osteoporosis mechanism.Methods Using the TCMSP database to search for the chemical components and targets of Busui mini-pills,and searching for genes related to osteoporosis through multiple databases such as DrugBank,GeneCards,Disgenet,TTD,and OMIM,Cytoscape 3.2.1 was used to construct compound-target network and protein interaction(PPI)network to screen out the core targets.Finally,GO function enrichment analysis and KEGG pathway enrichment analysis was performed through MetaScape to study the mechanism of action.Results 78 compounds with 307 corresponding protein targets were screened from Busui mini-pills.PPI network screened out 166 core targets including MAPK1,TP53,ESR1 and AKT1,which mainly involved cancer signaling pathways,MAPK signaling pathway,apoptosis signaling pathway and PI3K-Akt signaling pathway,etc.Conclusion Busui mini-pills are multi-component,multi-target,and overall regulation to treat osteoporosis.The potential mechanism of Busui mini-pills against osteoporosis was explored through network pharmacology,which provides a theory for its active ingredient research and further clinical experimental research.
