摘要
[目的]探索小金钗春石斛原球茎增殖的最佳培养基组分及试管苗移栽的适宜条件,为提高春石斛工厂化生产效率提供参考。[方法]采用L_(9)(3^(4))正交试验探讨6-BA、IBA和椰汁对原球茎增殖的影响;采用三因素两水平L_(4)(2^(3))的完全随机设计,筛选春石斛试管苗移栽最适条件。[结果]小金钗春石斛原球茎增殖最适培养基为:KC+1.5 mg·L^(-1)6-BA+0.75 mg·L^(-1) IBA+100 mL·L^(-1)椰汁+20 g·L^(-1)白糖+5.5 g·L^(-1)琼脂,pH 5.6。影响原球茎增殖效果最显著的是6-BA,其次是IBA,椰汁的影响最小。试管苗移栽最适条件为遮光、未消毒和选择较大的苗。试管苗大小、是否消毒及是否遮光对移栽效果均有极显著影响。[结论]6-BA是小金钗春石斛原球茎增殖的最主要影响因子;试管苗移栽最适条件为选择较大的试管苗移栽之后不用消毒处理,并进行遮光培养。
[Purpose]The optimum medium composition,a vital factor for the proliferation and transplanting of protocorm-like bodies of Dendrboium nobile,were investigated in the study,to provide guiding information for the industrialized cultivation of D.nobile with higher efficiency.[Method]Effect of 6-BA,IBA and coconut juice on the proliferation of the protocorm-like bodies of D.nobile were assessed by L_(9)(3^(4))orthogonal experimental design,and the optimum transplanting condition of D.nobile seedlings in vitro was analyzed by complete randomized design L_(4)(2^(3)).[Result]The optimum medium for the proliferation of D.nobile protocorm-like bodies was the Knudson C medium(pH is 5.6)supplemented with 1.5 mg·L^(-1)6-BA,0.75 mg·L^(-1) IBA,100 mL·L^(-1) coconut water,20 g·L^(-1) white sugar and 5.5 g·L^(-1) agar.6-BA was the primary factor for enhanced proliferation,followed by IBA and coconut water in a descending order.Shading condition,sterile condition and seedling size all significantly affected the transplanting outcome of D.nobile seedling,and thus bigger seedling was suggested during transplanting and cultivated under unsterilized and shading conditions later.[Conclusion]6-BA was the key factor for the proliferation of D.nobile protocorm-like bodies;larger seedling was recommended for transplanting,unsterilized and kept in shading conditiion.
