新型异黄酮对高糖诱导的白内障防治作用

摘要 目的探讨新型异黄酮对高糖诱导的白内障防治作用。方法8周龄雄性Wistar大鼠随机分为正常组(10只,给予0.3%羧甲基纤维素溶液),对照组(10只,给予10%高浓度果糖溶液),异黄酮低剂量组(9只,给予新型异黄酮1.0 mg·kg^(-1)·d^(-1))和异黄酮高剂量组(9只,给予新型异黄酮10 mg·kg^(-1)·d^(-1))。采用血压仪检测大鼠收缩压和舒张压,血糖仪监测大鼠血糖,眼底镜检查大鼠视觉。照相法记录大鼠晶状体的形态改变。取大鼠晶状体,采用Ellman法测定还原型谷胱甘肽(GSH);过氧化氢分解法测定晶状体过氧化氢酶(CAT)水平;硫代巴比妥酸反应来估计脂质过氧化(MDA)水平;比色法测定谷胱甘肽过氧化物酶(GSH-Px)。硝基蓝四唑还原法测定超氧化物歧化酶(SOD)。结果从第4周开始,对照组的收缩压、舒张压、血糖水平较正常组上升(均P<0.05),异黄酮低剂量组和异黄酮高剂量较对照组的收缩压、舒张压、血糖水平降低(均P<0.05)。4组大鼠第8周末形成白内障,与对照组相比,异黄酮低剂量和异黄酮高剂量组降低了3期和4期白内障的发生率(P<0.05,P<0.01)。与对照组相比,异黄酮低剂量组和异黄酮高剂量组的晶状体混浊情况明显好转(P<0.05)。异黄酮低剂量组和高剂量组的GSH、GSH-Px、CAT、SOD和MDA的水平高于对照组(P<0.05)。结论新型异黄酮可能通过降低高血压和糖尿病患者的晶状体氧化应激来预防白内障的形成。 Objective To investigate the effects of novel Isoflavones on the prevention and treatment of high glucose-induced cataracts.Methods Eight-week-old male Wistar rats were randomly divided into the normal group(n=10,given 0.3%Carboxymethyl cellulose),control group(n=10,given 10%high-concentration fructose),low-dose Isoflavones group(n=9,given novel Isoflavones 1.0 mg·kg^(-1)·d^(-1))and high-dose Isoflavones group(n=9,given novel Isoflavones 10 mg·kg^(-1)·d^(-1)).The systolic and diastolic blood pressures were determined by blood pressure meter.The blood glucose was determined by blood glucose meter,and the vision was examined by ophthalmoscope.The morphological changes of the lens were recorded by photography.The rat lenses were taken,and the Ellman assay,hydrogen peroxide decomposition method,thiobarbituric acid reaction,colorimetric determination,and nitro blue tetrazole reduction method were used to measure the levels of reduced glutathione(GSH),lens catalase(CAT),lipid peroxidation(MDA),glutathione peroxidase(GSH-Px),and superoxide dismutase(SOD),respectively.Results Starting from the 4th week,the systolic blood pressure,diastolic blood pressure,and blood glucose levels in the control group significantly increased compared with those in the normal group(all P<0.05).Compared with the control group,the systolic blood pressure,diastolic blood pressure,and blood glucose levels significantly decreased in the low-dose and high-dose Isoflavones groups(all P<0.05).The 4 groups of rats developed cataracts at the end of 8 week.Compared with the control group,the incidence of stage 3 and 4 cataract formation was significantly reduced in the low-dose and high-dose Isoflavones groups,respectively(P<0.05,P<0.01).Compared with the control group,the lens opacity was significantly improved in the low-dose and high-dose Isoflavones groups(all P<0.05).The levels of GSH,GSH-Px,CAT,SOD and MDA in the low-dose and high-dose Isoflavones groups were significantly higher than those in the control group(all P<0.05).Conclusion The novel Isoflavones may prevent the formation of cataracts by reducing the lens oxidative stress in patients with hypertension and diabetes.
