摘要
目的:通过比较两种混合造模对虚证小鼠体征及cAMP/cGMP、AQP4的影响,探寻综合性虚损症模型小鼠造模方法。方法:将KM小鼠随机分为正常对照组、模型组A、模型组B,进行灌胃番泻叶水浸液、游泳、失血造模,每周检测体质量、自主活动、体温、游泳力竭时间,连续3周。处死动物,检测血浆cAMP、cGMP含量,脏器指数和肾脏组织AQP4蛋白表达。结果:模型组A、模型组B均能明显地造成小鼠体质量增长变慢,体温下降,自主活动减少,游泳力竭时间缩短,血浆cAMP、cGMP含量升高,AQP4蛋白表达降低,且模型组B还使肝、脾、肾指数增加。结论:模型组A、模型组B在造模2周后能造成小鼠虚证模型,造模3周特征表现更明显。模型组B比模型组A损伤更明显。
Objective:To explore the optimal modeling methods of comprehensive deficiency syndrome by comparing the effects of two mixed modeling methods on signs,cAMP/cGMP and AQP4 in mice.Methods:KM mice were randomly divided into the normal group,the model group A and the model group B.The mice were established comprehensive deficiency syndrome by being fed with senna leaf water extract,swimming exhaustion and blood loss.Body weight,autonomous activities,body temperature and swimming exhaustion time were measured weekly for 3 weeks.The mice were sacrificed and the plasma levels of cAMP and cGMP,the organ indexes and the protein expression of AQP4 in renal tissues were detected in the groups.Results:The growth of body weight were slowed down,the body temperature was decreased,the autonomous activities were reduced,the exhaustive swimming time was shortened,the plasma contents of cAMP and cGMP were increased,and the protein expression of AQP4 was decreased in the model group A and the model group B;the indexes of liver,spleen and kidney were increased in the model group B.Conclusion:The deficiency syndrome model was induced after 2 weeks of modeling in the model group A and the model group B.The characteristics of the model became more obvious after 3 weeks of modeling in the two groups.The damages in the model group B were more significant than those in the model group A.
