摘要
采用HPLC-DAD对菊芋叶片中绿原酸的含量进行了测试,并考察了乙醇体积分数、料液比、提取温度和提取时间对菊芋叶片中绿原酸提取率的影响.通过正交实验确定的绿原酸最佳提取工艺条件为:乙醇体积分数70%、料液比1∶25、提取温度80℃、提取时间90 min.在最佳提取工艺条件下,对2020年菊芋生长周期内的绿原酸动态含量进行了分析,结果显示2020年11月份的菊芋叶片中绿原酸的含量最高,为1.52%(质量分数).本研究可为菊芋叶片中绿原酸的提取提供一定的参考,同时也扩大了富含绿原酸原料的来源,可为绿原酸的大规模开发和利用提供数据支撑.
The content of chlorogenic acid(CA)in Helianthus tuberosus leaves was determined by HPLC-DAD,and the effects of ethanol volume fraction,ratio of material to liquid,extraction temperature and extraction time on the extraction rate of CA from H.tuberosus leaves were investigated.The optimum extraction conditions of CA by orthogonal experiment were as follows:ethanol volume fraction 70%,material to liquid ratio 1∶25,extraction temperature 80℃,extraction time 90 min.Dynamic content of CA in H.tuberosus leaves in different growth stages was achieved.The content of CA in H.tuberosus leaves was the highest(1.52%,mass fraction)in November.This study can provide a certain reference for the extraction of CA from H.tuberosus leaves,and also expand the source of raw materials rich in CA,which can provide data support for the large-scale development and utilization of CA.
