摘要
目的:建立高效液相色谱(HPLC)法同时测定翻白草药材中4种黄酮类成分的方法,以及紫外-可见分光光度法测定翻白草总黄酮含量的方法,并比较19批翻白草药材和2批委陵菜药材上述成分含量差异。方法:采用HPLC法,色谱柱:InertSustain C_(18)(150 mm×4.6 mm,5μm),以0.1%甲酸溶液-乙腈为流动相梯度洗脱,流速1.0 mL/min,柱温30℃,检测波长360 nm,测定不同批次翻白草药材和委陵菜药材中4种黄酮类成分含量;以芦丁为对照品,采用紫外-可见分光光度法测定总黄酮的含量。结果:不同批次翻白草药材中4种黄酮类成分差异显著,总黄酮存在一定差异;委陵菜药材中芦丁、槲皮苷含量低于翻白草药材。结论:本研究所建立的方法稳定,精密度好,重现性高,为更好地控制翻白草药材的内在质量提供了科学依据。
Objective:To establish a high performance liquid chromatography(HPLC)method for simultaneous determination of four flavonoids in the herbal materials of Fanbaicao(Potentillae Discoloris Herba),and a ultraviolet-visible spectrophotometry to determine the content of total flavonoids in Fanbaicao,to compare the content differences between 19 batches of Fanbaicao and 2 batches of Weilingcai(Potentillae Chinensis Herba).Methods:HPLC method analysis was carried out on InertSustain C_(18) column(150 mm×4.6 mm,5μm)with 0.1%methanoic acid-acetonitrile as mobile phase in a gradient elution mode.The flow rate was 1.0 mL/min,the column temperature was 30℃,and the detection wavelength was set at 360 nm,which was used to determine the four flavonoids ingredients in samples.The content of total flavonoids was determined by ultraviolet-visible spectrophotometry with rutin as reference substance.Results:There were significant differences of the four flavonoids ingredients in 19 batches of Fanbaicao,and total flavonoids has some differences.The content of rutin and quercitrin in Weilingcai is far lower than that in Fanbaicao.Conclusion:The method established in this study is stable,accurate and reproducible,which provides a scientific basis for better control of the internal quality of Fanbaicao.
