荧光光谱法研究琥珀酸美托洛尔和壳聚糖之间的相互作用

摘要 目的:研究琥珀酸美托洛尔和壳聚糖之间的相互作用的基本原理,为壳聚糖作为美托洛尔药物制剂辅料作为参考。方法:用荧光光谱法考察了体系pH值、缓冲液用量、水浴时间、水浴温度、壳聚糖浓度对荧光猝灭的影响,测定了不同类型壳聚糖与琥珀酸美托洛尔之间的Stem-Volmer猝灭常数K sv、结合常数和结合位点数。用差示扫描量热法和X衍射法对复合物进行了表征。结果:药物与壳聚糖在pH值3.57的BR缓冲液中,温度为298 K时的结合比例为1∶1,结合常数为8.28×10^(5) L·mol^(-1),其猝灭类型属于静态猝灭,其作用力以氢键为主。结论:选择不同的壳聚糖的分子量,琥珀酸美托洛尔类药物制剂的释放效果不同。 Objective:To study the basic principle of the interaction between metoprolol succinate and chitosan,chitosan is used as an excipient for metoprolol pharmaceutical preparations as a reference.Methods:The effects of pH value,buffer dosage,water bath time,water bath temperature,and chitosan concentration on fluorescence quenching were investigated by fluorescence spectroscopy,and the Stem-between different types of chitosan and metoprolol succinate was determined.Volmer quenching constant K sv,binding constant and number of binding sites.The composite was characterized by differential scanning calorimetry and X-ray diffraction method.Results:The binding ratio of the drug and chitosan in BR buffer at pH 3.57 at a temperature of 298 K is 1∶1,and the binding constant is 8.28×10^(5) L·mol^(-1),which quenches The type belongs to static quenching,and its force is dominated by hydrogen bonds.Conclusion:According to the choice of different molecular weight of chitosan,the release effect of metoprolol succinate preparations is different.
