摘要
目的探讨Yes相关蛋白1(YAP1)可能通过调控mTOR/p70s6k信号通路对卵巢癌细胞生长和转移的影响。方法以卵巢癌细胞SKOV3为研究对象,将细胞分为si-NC组(转染si-NC)、si-YAP1组(转染si-YAP1)和si-YAP1+3-BDO组(转染si-YAP1后,加入100μmol/L的mTOR通路激活剂3-BDO)。然后采用qRT-PCR检测YAP1 mRNA的表达量;CCK8检测细胞活力;Western blot检测YAP1、Ki67、E-cadherin、N-cadherin和mTOR/p70s6k通路相关蛋白的表达;Transwell检测细胞迁移和侵袭。结果si-YAP1组的YAP1 mRNA和蛋白表达、细胞活力、Ki67蛋白表达、迁移细胞数目、侵袭细胞数目、N-cadherin蛋白表达、p-mTOR蛋白表达、p-p70s6k蛋白表达明显低于si-NC组,E-cadherin蛋白表达明显高于si-NC组。si-YAP1+3-BDO组的p-mTOR蛋白表达、p-p70s6k蛋白表达、细胞活力、Ki67蛋白表达、迁移细胞数目、侵袭细胞数目、N-cadherin蛋白表达明显高于si-YAP1组,E-cadherin蛋白表达明显低于si-YAP1组。结论沉默YAP1可能通过激活mTOR/p70s6k信号通路促进卵巢癌细胞的增殖、迁移和侵袭。
Objective To investigate the effect of Yes-related protein 1(YAP1)on the growth and metastasis of ovarian cancer cells,focusing on the regulation of the mTOR/p70s6k signaling pathway.Methods The ovarian cancer SKOV3 cells were divided into si-NC group(transfected si-NC),si-YAP1 group(transfected si-YAP1),and si-YAP1+3BDO group(after transfection of si-YAP1,100μmol/L mTOR pathway activator 3BDO was added).Then qRT-PCR was used to detect the expression of YAP1 mRNA;CCK8 to detect cell viability;Western blot to detect the expression of YAP1,Ki67,E-cadherin,N-cadherin and mTOR/p70s6k pathway related proteins;Transwell to detect cell migration and invasion.Results The YAP1 mRNA and protein expression,cell viability,Ki67 protein expression,number of migrating cells,number of invasive cells,N-cadherin protein expression,p-mTOR protein expression,and p-p70s6k protein expression in the si-YAP1 group were significantly lower than those in the si-NC group;E-cadherin protein expression was significantly higher than that of si-NC group.The p-mTOR protein expression,p-p70s6k protein expression,cell viability,Ki67 protein expression,number of migrating cells,number of invasion cells,and N-cadherin protein expression in the si-YAP1+3BDO group were significantly higher than those in the si-YAP1 group;and the expression of E-cadherin protein was significantly lower than that of si-YAP1 group.Conclusion Silencing YAP1 may promote the proliferation,migration and invasion of ovarian cancer cells by activating the mTOR/p70s6k signaling pathway.
