基于指纹图谱及多成分含量测定的黔产金钗石斛花质量控制研究 被引量:1

摘要 目的建立黔产金钗石斛花高效液相色谱(HPLC)的指纹图谱、总黄酮及多成分含量测定方法。方法采用Agilent Eclipse Plus C_(18)色谱柱(4.6 mm×150 mm,5μm);以乙腈(A)-0.06%醋酸-水溶液(B)为流动相,梯度洗脱;流速1.0 mL/min;检测波长280 nm;柱温30℃。结果14批金钗石斛花指纹图谱相似度均大于0.954,共标定13个共有峰,指认了甘草苷、山柰酚-3-葡萄糖鼠李糖苷和异槲皮苷3个成分,其含量分别为1.4407~3.2287、5.9434~10.1619和1.2188~2.9090 mg/g。14批样品的总黄酮含量为0.054~0.082 g/g。聚类分析(HCA)发现冻干和烘干样品存在差异。结论本研究所建立的HPLC多成分含量测定和指纹图谱的方法准确可行,可作为黔产金钗石斛花质量评价的一种有效手段。 Objective To establish a HPLC method for fingerprint and multi-component determination of Dendrobium nobile Lindl.flower.Methods Agilent Eclipse Plus C_(18) column(4.6 mm×150 mm,5μm)was used.With acetonitrile(A)-0.06%acetic acid-water solution(B)as mobile phase,gradient elution was performed.The flow rate was 1.0 mL/min.The detection wavelength was 280 nm.The column temperature was 30℃.Results The similarity of 14 batches of Dendrobium nobile Lindl.flower fingerprints were all greater than 0.954.A total of 13 common peaks were identified,and 3 components including liquiritin,kaempferol-3-glucorate rhamnoside and isoquercitrin were identified,the contents were 1.4407~3.2287,5.9434~10.1619 and 1.2188~2.9090 mg/g respectively.The total flavonoids content of 14 batches of samples ranged from 0.054 to 0.082 g/g.Cluster analysis(HCA)showed that there were differences between lyophilized and dried samples.Conclusion The method established by this research is accurate and feasible,and can be used as an effective means for quality evaluation of Dendrobium nobile Lindl.flower.
