中空Gd_(2)O_(3)微球的制备与表征及其在小鼠模型成像中的应用

摘要 研发高性能T_(1)型核磁共振造影剂仍是目前重要的科研任务.文中首先以葡萄糖为单元,利用水热法制备了碳微球,并以此为球模板,结合高温热解技术,成功制备了中空Gd_(2)O_(3)微球.利用傅立叶变换红外光谱、热重、X射线衍射、X射线光电子能谱、扫描电子显微镜和透射电子显微镜等对其结构进行了表征.结果表明,中空Gd_(2)O_(3)微球的直径约为500 nm,壳厚约为20 nm.细胞实验表明所制备中空Gd_(2)O_(3)微球体外毒性低;体外核磁共振成像显示该微球的纵向和横向弛豫率分别为10.46和13.83 L·mmol^(-1)·s^(-1),具有较高的T1弛豫率;小鼠模型成像结果表明该中空微球可提高小鼠主要组织图像的亮度,表现出良好的应用前景. The research of high-performance T_(1) MRI contrast agent is still an important scientific research task.In this work,firstly,carbon microspheres were prepared by hydrothermal method with glucose as unit,and hollow Gd_(2)O_(3) microspheres were successfully prepared by using this as ball template and high temperature pyrolysis technology.The structure of hollow Gd_(2)O_(3) microspheres were characterized by Fourier transform infrared spectroscopy,thermogravimetric analysis,X-ray diffraction spectroscopy,Xray photoelectron spectroscopy,scanning electron microscopy and transmission electron microscopy.The results showed that the diameter of hollow Gd_(2)O_(3) microspheres are about 500 nm and the shell thickness is about 20 nm.Cell experiments showed that the prepared hollow Gd_(2)O_(3) microspheres has low toxicity in vitro.In vitro nuclear magnetic resonance imaging showed that the longitudinal and transverse relaxation rates of the microspheres are 10.46 and 13.83 L·mmol^(-1)·s^(-1),respectively,with high T_(1) relaxation rates.The imaging results of mouse model showed that the hollow microspheres can improve the brightness of main tissue images of mice,showing a good application prospect.
