大鼠单侧耳蜗损伤后下丘中GluR2/3受体的变化

摘要 目的观察单侧耳蜗损伤后短时间内谷氨酸受体2/3(glutamate receptor 2 and 3,GluR2/3受体)在大鼠下丘的表达变化,为探讨外周损伤后听觉中枢重组的分子机制提供参考。方法36只大鼠随机分为9组,分别为耳蜗损伤造模后2 h、4 h、6 h、8 h、12 h、24 h、3 d、7 d及正常对照组,每组4只,前8组动物用手术方法制造单侧耳蜗损伤模型,正常对照组不做处理,造模完成后于相应时间点采用FITC标记免疫组化方法检测各组GluR2/3受体在下丘的表达变化。结果GluR2/3受体主要位于胞膜及膜周胞浆中,胞核周弱表达或无表达;造模术后不同时间各组术侧下丘中GluR2/3受体相对灰值比较,术侧与健侧比、手术组与对照组比较,差异均无统计学意义(均为P>0.05)。结论大鼠单侧耳蜗损伤7天内术侧下丘中GluR2/3表达未见明显差异。
