槐米提取物对衰老模型小鼠运动能力的影响及机制研究 被引量:4

摘要 目的探讨槐米提取物对D-半乳糖致衰老模型小鼠运动能力的影响及相关机制。方法采用小鼠颈背部皮下注射D-半乳糖(500 mg·kg^(-1))建立衰老小鼠模型,每日1次,连续9周。将48只小鼠随机分为6组:空白组、模型组、维生素C组(200 mg·kg^(-1))和槐米提取物低、中、高剂量组(100、200、400 mg·kg^(-1)),造模第5周开始,同时灌胃相应药物(10 mL·kg^(-1)),每日1次,连续4周。观察小鼠的行为变化并定时记录小鼠体质量;通过行为学疲劳实验和负重游泳实验考察小鼠的运动能力;测定小鼠胸腺、脾脏及脑组织的脏器指数;检测血清及脑组织中的总超氧化物歧化酶(T-SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性和丙二醛(MDA)含量;采用HE染色法观察小鼠脑组织病理变化。结果与空白组比较,模型组小鼠的在棒时间及游泳时间显著缩短(P<0.01);小鼠胸腺、脾脏及脑指数明显降低(P<0.05,P<0.01);小鼠血清及脑组织中MDA含量明显增高(P<0.05,P<0.01),T-SOD、GSH-Px活性明显降低(P<0.05,P<0.01)。与模型组比较,槐米提取物中剂量组小鼠的在棒时间和游泳时间明显延长(P<0.05),小鼠胸腺指数明显升高(P<0.05);槐米提取物中、低剂量组小鼠的血清T-SOD、GSH-Px活性显著升高(P<0.01),脑组织中T-SOD活性显著升高(P<0.01),血清及脑组织中的MDA含量显著降低(P<0.01)。结论槐米提取物可提高衰老模型小鼠的运动能力,其作用机制可能与提高机体内抗氧化酶活性、降低脂质过氧化有关。 Objective To investigate the effects and mechanism of Sophorae flos extract on exercise performance of D-galactose induced aging model mice.Methods Mice were subcutaneously injected with D-galactose(500 mg·kg^(-1))into the napes once a day for consecutive 9 weeks,to establish the aging mouse model.48 successfully modeled mice were randomly divided into blank group,model group,VC group,and low,medium,high doses of Sophorae flos extract groups(100,200,and 1400 mg · kg^(-1)).Drugs were intragastrically administered once daily for4 consecutive weeks.Changes in behavior and body mass were observed regularly.The behavioral fatigue test and weight-bearing swimming test were conducted after the last administration.The thymus,spleen and brain indexes of the mice were measured,and the activities of T-SOD,GSH-Px and content of MDA in serum and brain tissues were determined.The pathological morphological changes of brain of the mice were observed by HE staining.Results Compared with the blank group,mice in the model group had significantly shorter rod time and swimming time(P<0.01).The thymus,spleen and brain indexes of the model group were significantly decreased(P<0.05,P<0.01).The MDA contents in serum and brain tissue were significantly increased(P<0.05,P<0.01),while the activities of T-SOD and GSH-Px were significantly decreased(P<0.05,P<0.01).Compared with the model group,the mice in the medium dose Sophorae flos extract group had significantly longer rod time and swimming time(P<0.05);the indexes of thymus of mice were increased(P<0.05).For mice in medium and low dose Sophorae flos extract groups,the activities of T-SOD and GSH-Px in serum and T-SOD activities in brain tissues were increased(P<0.01),and the levels of MDA in serum and brain tissue were decreased(P<0.01).Conclusion Sophorae flos extract can improve exercise performance of subacute aging model mice,its mechanism may be related to improve the activities of antioxidant enzymes and reduce lipid peroxidation.
