培养基组分及培养条件对米曲霉麦角甾醇含量的影响 被引量:2

摘要 本文建立了简便的米曲霉麦角甾醇含量的检测方法,并研究不同培养条件对其麦角甾醇含量的影响。通过醇碱皂化法提取米曲霉中的麦角甾醇再经过高效液相色谱法定量分析来研究不同培养条件对米曲霉合成麦角甾醇量的影响。研究发现:麦角甾醇合成量随着米曲霉生长时期的增长而增加;适当高温会使甾醇含量增加,而温度过高和过低都会降低麦角甾醇的合成量;在NaCl和乙醇胁迫下,低浓度不会对麦角甾醇合成有显著影响,而高浓度则会明显降低麦角甾醇的积累甚至影响米曲霉的正常生长;培养基的成分对米曲霉合成麦角甾醇也具有显著性影响。结果表明不同的培养条件和培养基组分对米曲霉中麦角甾醇合成量有影响,对培养条件和培养基组分进行优化可以提高米曲霉产麦角甾醇的量。 This article established a simple method for detecting ergosterol content of A.oryzae and studied the influence of different culture conditions on its ergosterol content.Ergosterol was extracted from A.oryzae by alcoholbase saponification and determined by high performance liquid chromatography(HPLC) to reveal the effect of different culture conditions on the content of ergosterol in A.oryzae.The concentration of ergosterol increases with the growth of A.oryzae;appropriate high temperature increases the sterol content;Low concentrations of NaCl and ethanol stress do not significantly affect ergosterol concentration,while high concentrations of NaCl and ethanol stress significantly reduce the accumulation of ergosterol;the composition of the medium also has a significant effect on the synthesis of ergosterol by A.oryzae.The results show that different culture conditions and medium components have an effect on the synthesis of ergosterol in A.oryzae.Optimizing the culture conditions and medium components can increase the amount of ergosterol produced by A.oryzae.
