摘要
为研究苯并芘对鱼类外周血红细胞及抗氧化酶系的影响,本文以泥鳅作为研究对象,将其暴露于0、5、10、15μg/LBaP溶液中20 d,分别在5、10、15、20 d时取样,采用血涂片微核测定法观察红细胞微核与核异常,采用邻苯三酚自氧化法和钼酸铵比色法分别检测肝脏超氧化物歧化酶(Superoxide Dismutase, SOD)和过氧化氢酶(Catalase,CAT)活性。结果表明,随着BaP浓度的升高和暴露时间的延长,泥鳅血红细胞微核率显著升高;随着BaP浓度的升高,泥鳅SOD、CAT活性在20 d的暴露期内呈现出先升高后下降的趋势。因此,BaP胁迫会对泥鳅产生一定的遗传毒性和氧化毒性效应,且毒性作用具有时间-剂量效应。
To study the effect of Benzo(a)pyrene on erythrocyte and antioxidant enzyme activities in fish, loach was exposed to 0,5,10 and 15 μg/L BaP solution for 20 days. During the BaP treatment, the blood and liver were collected on 5,10,15 and 20 days. The erythrocyte micronucleus and nuclear abnormality were detected by blood smear, the activities of superoxide dismutase(SOD) and catalase(CAT) in liver were measured by pyrogallol autoxidation and ammonium molybdate colorimetry. The results showed that:(1) With the increase of BaP concentration and exposure time, the micronucleus rate of erythrocytes of M. anguillicaudatus increased significantly.(2) With the increase of BaP concentration, the activities of SOD and CAT of M. anguillicaudatus increased at first and then decreased during 20 days of exposure. Therfore, BaP stress could induce certain genotoxicity and oxidative toxicity to Misgurnus anguillicaudatus, and the toxicity had time-dose effect.
