实验树鼩体内卵泡抑素样蛋白1表达与室性心动过速的相关性分析 被引量:1

摘要 目的探究实验动物体内卵泡抑素样蛋白1(follistatin like protein 1,FSTL1)表达与室性心动过速(室速)的相关性。方法通过药物诱发成功构建实验树鼩室速模型40只,根据随机数表法分为对照组、室速1 h组、室速2 h组、室速3 h组,每组10只。采用ELISA、RT-qPCR、Western blot以及免疫组织化学法检测各组FSTL1表达。结果室速1 h组、室速2 h组、室速3 h组ELISA检测外周血FSTL1表达较对照组明显增加[(20.61±7.28)ng/ml、(34.85±6.49)ng/ml、(19.97±8.74)ng/ml vs(12.47±5.52)ng/ml,P<0.05]。室速2 h组外周血FSTL1表达较室速1 h组明显增加,差异有统计学意义(P<0.01)。室速3 h组外周血FSTL1表达较室速2 h组明显降低,差异有统计学意义(P<0.05)。室速1 h组、室速2 h组、室速3 h组心肌组织FSTL1 mRNA和FSTL1蛋白表达较对照组明显增加,差异有统计学意义(1.46±0.32、1.87±0.44、1.36±0.43 vs 1.00±0.26,1.48±0.36、1.94±0.47、1.46±0.41 vs 1.00±0.33,P<0.05,P<0.01)。室速2 h组心肌组织FSTL1 mRNA和蛋白表达最高,较室速1 h组和室速3 h组FSTL1 mRNA和蛋白表达明显增加,差异有统计学意义(P<0.05)。免疫组织化学检测显示,对照组FSTL1表达为阴性或弱阳性,室速1 h组FSTL1表达为阳性,室速2 h组FSTL1表达为强阳性,阳性强度最高;室速3 h组阳性FSTL1表达较室速2 h组降低。结论实验树鼩FSTL1蛋白表达量随着室速持续时间的延长表现为先升高后回落的趋势。 Objective To study the correlation between follistatin like protein 1(FSTL1)expres-sion and ventricular tachycardia(VT)in laboratory tree shrews.Methods A drug-induced VT model of laboratory tree shrews was established.Forty tree shrews were randomly divided into control group,1 h VT group,2 h VT group,and 3 h VT group(10 in each group).FSTL1 expres-sion was detected by ELISA,RT-qPCR,Western blot respectively with immunohistochemical staining.Results The expression level of FSTL1 in peripheral blood was significantly higher in 1 h,2 h and 3 h VT groups than in control group(20.61±7.28 ng/ml,34.85±6.49 ng/ml,19.97±8.74 ng/ml vs 12.47±5.52 ng/ml,P<0.05)and in 2 h VT group than in 1 h VT group(P<0.01),and was significantly lower in 3 h VT group than in 2 h VT group(P<0.05).The exres-sion levels of FSTL1 mRNA and protein in myocardial tissue were significantly higher in 1 h,2 h and 3 h VT groups than in control group(1.46±0.32,1.87±0.44,1.36±0.43 vs 1.00±0.26;1.48±0.36,1.94±0.47,1.46±0.41 vs 1.00±0.33,P<0.05,P<0.01)and in 2 h VT group than in 1 h and 3 h VT groups(P<0.05).Immunohistochemical staining showed that FSTL1 was negatively and weakly expressed in control group,positively expressed in 1 h VT group,high-ly and positively expressed in 3 h VT group.The positive expression level of FSTL1 was signifi-cantly lower in 3 h VT group than in 2 h VT group.Conclusion The FSTL1 protein expression increases at first and then decreases with the prolonged time of VT in laboratory tree shrews.
