摘要
为了解中国东北虎源猫免疫缺陷病毒(FIV)的感染情况,采集海林市横道河子东北虎林园东北虎全血样本50份,通过套式PCR对样本进行FIV pol基因扩增,并对阳性样本进行测序,再运用生物信息学软件进行遗传进化分析。结果检测出1只虎为FIV阳性,其pol基因与参考毒株的核苷酸和氨基酸序列同源性分别为70.0%~99.8%和75.0%~99.4%,且存在39个氨基酸突变位点,系统发育树显示其与A亚型猫源FIV CHN17毒株亲缘关系最近。这是中国首次从东北虎体内检测到FIV,表明对中国大型猫科动物进行FIV的监测的必要性,同时应该注意对圈养老虎栖息地周围流浪猫的管理。
In order to understand the infection status of feline immunodeficiency virus(FIV)from Siberian tigers in China,50 whole blood samples of Siberian tigers were collected from Siberian Tiger Park in Hengdaohezi,Hailin city,and the FIV pol gene was amplified by nested PCR,and then phylogenetic analysis was performed.As a result,one sample was FIV positive.The nucleotide and amino acid sequence homologies between the pol gene with the references were 70.0%-99.8%and 75.0%-99.4%,respectively,and there were 39 amino acid mutation sites.The phylogenetic tree shows that it is closely related to CHN17 strain,which is subtype A FIV from cat.This is the first time FIV from a Siberian tiger was detected in China,so it is necessary to monitor FIV of Chinese wild felines,and more attention should be paid to the management of stray cats around captive tiger habitats.
