miR-182-5p、TRIM8在子宫内膜癌组织中的表达及其临床意义

Expression and clinical significance of miR-182-5p and TRIM8 in endometrial cancer

目的探讨miRNA-182-5p(miR-182-5p)和三结构域蛋白8(TRIM8)在子宫内膜癌组织中的表达及其临床意义。方法选取2015年1月—2016年6月于徐州市中心医院行手术治疗的子宫内膜癌患者76例,收集手术切除的子宫内膜癌组织及癌旁正常子宫内膜组织。采用免疫组化法检测组织中TRIM8蛋白表达水平;qRT-PCR法检测miR-182-5p、TRIM8 mRNA表达水平;Spearman法进行miR-182-5p与TRIM8的相关性分析;Kaplan-Meier法及Cox比例风险回归模型分析子宫内膜癌患者预后的影响因素。结果免疫组化结果显示,子宫内膜癌组织中TRIM8蛋白阳性表达率(55.26%)低于癌旁组织(73.68%)(P<0.05)。qRT-PCR检测结果显示,子宫内膜癌组织中miR-182-5p的表达水平显著高于癌旁组织,而TRIM8的表达水平明显低于癌旁组织(P<0.05)。子宫内膜癌组织中miR-182-5p、TRIM8表达水平均与FIGO分期、淋巴结转移有关(P<0.05)。Spearman相关性分析显示,miR-182-5p与TRIM8表达水平呈负相关(r=-0.575,P<0.05)。Kaplan-Meier分析结果显示,miR-182-5p高表达组、TRIM8低表达组患者的预后较差(P<0.05)。Cox多因素回归分析显示,miR-182-5p、TRIM8是影响子宫内膜癌患者预后的独立危险因素。结论子宫内膜癌组织中miR-182-5p呈高表达,TRIM8呈低表达,二者的表达失调可能共同参与子宫内膜癌的发生发展。

Objective To investigate the expression of miRNA-182-5p(miR-182-5p)and three-domain protein 8(TRIM8)in endometrial cancer and its clinical significance.Methods A total of 76 patients with endometrial cancer who underwent surgical treatment in Xuzhou Central Hospital from January 2015 to June 2016 were selected.The surgically removed endometrial cancer tissues and adjacent normal tissues were collected.The expression of TRIM8 protein in the tissues was detected by immunohistochemistry.The levels of miR-182-5p and TRIM8 mRNA were detected by qRT-PCR.The correlation between miR-182-5p and TRIM8 was analyzed by Spearman method.The prognostic factors of endometrial cancer were analyzed by Kaplan-Meier method and Cox proportional risk regression model.Results Immunohistochemical results showed that the positive expression rate of TRIM8 in endometrial cancer tissues(55.26%)was lower than that in adjacent normal tissues(73.68%)(P<0.05).qRT-PCR results showed that endometrial cancer tissues showed increased miR-182-5p levels and decreased TRIM8 levels,compared with adjacent normal tissues(P<0.05).The expression of miR-182-5p and TRIM8 in endometrial cancer tissues was related with FIGO stage and lymph node metastasis(P<0.05).Spearman correlation analysis showed that miR-182-5p was negatively related with TRIM8 expression(r=-0.575,P<0.05).Kaplan-Meier analysis showed that patients with high miR-182-5 Pexpression and low TRIM8 expression had a poor prognosis(P<0.05).Cox multivariate regression analysis indicated that miR-182-5p and TRIM8 were the independent risk factors influencing the prognosis of patients with endometrial cancer.Conclusions The expression of miR-182-5p is up-regulated and the expression of TRIM8 is down-regulated in endometrial cancer tissue.The dysregulation of miR-182-5p and TRIM8 may be involved in the pathogenesis of endometrial cancer.
