摘要
应用引物2550F/2718R对上海动物园内蓝黄金刚鹦鹉和红绿金刚鹦鹉的CHD基因进行PCR扩增,进行性别鉴定。经过琼脂糖凝胶电泳发现:雄性个体扩增出1条片段,大小647 bp;雌性个体扩增出2条片段,大小分别为630和411 bp。蓝黄金刚鹦鹉群体的雌雄比为10∶17,红绿金刚鹦鹉群体的雌雄比为5∶4。该方法能准确、快速、有效地进行2种金刚鹦鹉的性别鉴定,为该鸟类种群饲养繁育提供基础性别信息。
Primers 2550F/2718R were used to amplify CHD gene of Ara ararauna and Ara chloropterus in Shanghai Zoo by PCR.Agarose gel electrophoresis results showed that the male individuals amplified one band with the length of 647 bp,while the female individuals amplified two bands with the length of 630 and 411 bp respectively.The male-female ratio of A.ararauna was 10∶17.The male-female ratio of A.chloropterus was 5∶4.This method could accurately,quickly and effectively identify the sex of two species of macaw,and provide the basic sex information for the feeding and breeding of macaw population.
