清肝化痰方对H型高血压大鼠TNF-α、IL-1β、IL-6表达水平及颈动脉结构的影响 被引量:2

Effects of Qinggan Huatan Recipe on Expression Levels of TNF-α,IL-1β,IL-6 and Carotid Artery Structure in H-type Hypertension Rats

摘要 目的探讨清肝化痰方对H型高血压大鼠肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)蛋白表达水平以及颈动脉结构的影响。方法将符合H型高血压模型的30只自发性高血压大鼠(SHR)随机分为模型组、马来酸依那普利叶酸(依叶)组、清肝化痰方组,每组10只。模型组:正常饲养8周;依叶组:正常饲养,以0.06%依那普利叶酸按10 mL/100 g灌胃,每日1次,连续干预8周;清肝化痰方组:正常饲养,加清肝化痰方,按成人体质量200 mL/60 kg(0.003 mL/g)浓度,2倍浓度灌胃,每日1次,连续干预8周。同源正常血压大鼠(WKY)10只作为对照组,不进行饲料添加处理。采用免疫组化技术测量大鼠颈动脉中膜厚度(CMT)、颈动脉内径(CAID)、颈动脉中膜横截面面积(CAMT);采用蛋白免疫印迹法检测大鼠主动脉中TNF-α、IL-1β、IL-6的蛋白表达情况;通过苏木精-伊红(HE)染色检测大鼠血管纤维化。结果清肝化痰方组、依叶组平均动脉压、舒张压、收缩压均较模型组降低(P<0.05或P<0.01),且清肝化痰方组平均动脉压、舒张压、收缩压均较依叶组明显降低(P<0.05)。依叶组和清肝化痰方组较模型组血管纤维化明显降低;与依叶组比较,清肝化痰方组血管纤维化明显降低。清肝化痰方组、依叶组CMT、CAMT较模型组明显降低(P<0.05或P<0.01),清肝化痰方组CMT、CAMT较依叶组明显降低(P<0.05);清肝化痰方组、依叶组CAID较模型组明显升高(P<0.05或P<0.01),清肝化痰方组CAID较依叶组明显升高(P<0.05)。依叶组TNF-α、IL-1β、IL-6蛋白表达水平较模型组降低(P<0.05),清肝化痰方组TNF-α、IL-1β、IL-6蛋白表达水平较模型组明显降低(P<0.01)。结论清肝化痰方通过降低H型高血压大鼠平均动脉压、舒张压、收缩压及TNF-α、IL-1β、IL-6蛋白表达水平,降低血管纤维化,一定程度上改善颈动脉异常变形。 Objective To investigate the effect of Qinggan Huatan Recipe on tumor necrosis factor(TNF-α),interleukin-1β(IL-1β),interleukin-6(IL-6)levels,and carotid artery structure in H-type hypertension rats.Methods Thirty spontaneous hypertensive rats(SHR)in H-type hypertension model were randomly divided into model group,enalapril folic acid group,Qinggan Huatan Recipe group,with 10 rats in each group.The rats in model group were fed normally for 8 weeks.On the basis of normal feeding,the rats in enalapril folic acid group received intragastric administration of 0.06%enalapril folic acid at 10 mL/100 g,once a day,for 8 weeks.The rats in Qinggan Huatan Recipe group received intragastric administration of Qinggan Huatan Recipe at twice the concentration of adult human body mass[200 mL/60 kg(0.003 mL/g)],once a day,for 8 weeks.Ten homologous Wistar-Kyoto(WKY)rats with normal blood pressure were used as control group without operation or feed addition.The carotid media thickness(CMT),carotid artery diameter(CAD),cross-sectional area of carotid media were measured by immunohistochemistry,and the protein expression of TNF-α,IL-1β,and IL-6 were detected by Western Blot.Results The mean arterial pressure,diastolic pressure,and systolic pressure in Qinggan Huatan Recipe group and enalapril folic acid group were lower than those in model group(P<0.05 or P<0.01),and the mean arterial pressure,diastolic pressure,and systolic pressure in Qinggan Huatan Recipe group were lower than those in enalapril folic acid group(P<0.05).Compared with model group,the cardiac fibrosis of enalapril folic acid group and Qinggan Huatan Recip group decreased.Compared with enalapril folic acid group,the cardiac fibrosis in Qinggan Huatan Recip group decreased.CMT and cross-sectional area of carotid media in Qinggan Huatan Recip group and enalapril folic acid group were lower than those in model group(P<0.05 or P<0.01),and CMT and cross-sectional area of carotid media in Qinggan Huatan Recip group were lower than those in enalapril folic acid group(P<0.05).CAD in Qinggan Huatan Recip group and enalapril folic acid group were higher than that in model group(P<0.05 or P<0.01),and CAD in Qinggan Huatan Recip group was higher than that in enalapril folic acid group(P<0.05).The expression levels of TNF-α,IL-1β,and IL-6 protein in enalapril folic acid group were lower than those in model group(P<0.05),and the expression levels of TNF-α,IL-1β,and IL-6 protein in Qinggan Huatan Recip group were lower than those in model group(P<0.01).Conclusion Qinggan Huatan Recipe can reduce mean arterial pressure,diastolic pressure,systolic pressure and the protein expression levels of TNF-α,IL-1β,and IL-6 in H-type hypertension rats,reduce cardiac fibrosis,and improve abnormal deformation of carotid artery.
