紫丁香苷对前交叉韧带横断诱导的大鼠骨关节炎的影响 被引量:8

摘要 目的探讨紫丁香苷对前交叉韧带横断(ACLT)诱导的骨关节炎大鼠病理损伤、炎症反应及Wnt/β-catenin信号通路的影响。方法将72只雄性SD大鼠随机分为假手术组、模型组、双醋瑞因组、紫丁香苷25 mg/kg组、紫丁香苷50 mg/kg组和紫丁香苷100 mg/kg组,每组12只,构建骨关节炎大鼠模型。苏木精—伊红(HE)染色和番红O-固绿染色观察关节软骨组织病理损伤程度,并进行Mankin's评分和国际骨关节炎研究学会(OARSI)半定量评分;ELISA检测血清和关节灌洗液中IL-6、TNF-α、IL-1β水平;Western blot检测MMP-13、Aggrecan、typeⅡcollagen、Wnt1、β-catenin蛋白表达水平。结果与假手术组比较,模型组出现明显骨关节炎关节软骨组织病理损伤,Mankin's评分、OARSI评分较高(P<0.05),血清和关节灌洗液中IL-6、TNF-α、IL-1β水平较高(P<0.05),MMP-13、Wnt1、β-catenin蛋白表达水平较高(P<0.05),Aggrecan、typeⅡcollagen蛋白表达水平较低(P<0.05);与模型组比较,双醋瑞因组、紫丁香苷50 mg/kg组和紫丁香苷100 mg/kg组骨关节炎关节软骨组织病理损伤明显改善,Mankin's评分、OARSI评分较低(P<0.05),血清和关节灌洗液中IL-6、TNF-α、IL-1β水平较低(P<0.05),MMP-13、Wnt1、β-catenin蛋白表达水平较低(P<0.05),Aggrecan、typeⅡcollagen蛋白表达水平较高(P<0.05)。结论紫丁香苷可通过抑制炎症反应、细胞外基质的降解及Wnt/β-catenin信号通路的活化改善ACLT诱导的骨关节炎大鼠关节软骨组织病理损伤。 Objective To investigate the effects of syringin on the pathological injury,inflammatory response and Wnt/β-catenin signaling pathway in rats with osteoarthritis induced by anterior cruciate ligament transection(ACLT).Methods A total of 72 male SD rats were randomly divided into the sham operation group,the model group,the diacerein group,the syringin 25 mg/kg group,the syringin 50 mg/kg group and the syringin 100 mg/kg group,with 12 rats in each group to construct the osteoarthritis rat model.The degree of pathological injury of articular cartilage tissue was observed by the hematoxylin-eosin(HE)staining and safranin O-fast green staining,and the Mankin's score and Osteoarthritis Research Society International(OARSI)score were performed.The levels of IL-6,TNF-αand IL-1βin serum and joint lavage fluid were detected by ELISA.The protein expression levels of MMP-13,Aggrecan,typeⅡcollagen,Wnt1 andβ-catenin were detected by Western blot.Results Compared with the sham operation group,the model group had obvious pathological injury of osteoarthritis articular cartilage tissue,with higher Mankin's score and OARSI score(P<0.05),higher levels of IL-6,TNF-αand IL-1βin serum and joint lavage fluid(P<0.05),higher protein expression levels of MMP-13,Wnt1 andβ-catenin(P<0.05),and lower protein expression levels of Aggrecan and typeⅡcollagen(P<0.05).Compared with the model group,the pathological injury of osteoarthritis articular cartilage tissue in the diacerein group,the syringin 50 mg/kg group and the syringin 100 mg/kg group were significantly improved,the Mankin's score and OARSI score were lower(P<0.05),the levels of IL-6,TNF-αand IL-1βin serum and joint lavage fluid were lower(P<0.05),the protein expression levels of MMP-13,Wnt1 andβ-catenin were lower(P<0.05),and the protein expression levels of Aggrecan and typeⅡcollagen were higher(P<0.05).Conclusion Syringin can improve the pathological injury of articular cartilage tissue in rats with osteoarthritis induced by ACLT by inhibiting inflammatory response,degradation of extracellular matrix and activation of Wnt/β-catenin signaling pathway.
