非肥胖糖尿病小鼠调节性B细胞与其他免疫细胞的关系

Relationship of regulatory B cells and other immune cells in non-obese diabetic mice

摘要 目的探讨非肥胖糖尿病(NOD)小鼠调节性B细胞(Breg细胞)与其他免疫细胞的关系。方法45只NOD小鼠均分为A组(4周龄)、B组(8周龄)和C组(1型糖尿病小鼠)三组。HE染色观察小鼠胰腺组织形态。流式细胞术检测三组小鼠脾脏调节性T细胞(Treg细胞)(CD4^(+)CD25^(+)Foxp3^(+))、Th1细胞(CD4^(+)IFN-γ^(+))、Th17细胞(CD4^(+)IL-17^(+)),B10细胞(CD19^(+)CD5^(+)CD1d^(hi)B)和IL-10^(+)B细胞(CD19^(+)IL-10^(+))数量。结果与A组比较,B组Th1细胞、Th17细胞、B10细胞和IL-10^(+)B细胞增多,而C组Treg细胞减少,Th1细胞、Th17细胞、B10细胞IL-10^(+)B细胞、Th1/B10比例和Th17/B10比例增多(P<0.05);与B组比较,C组Th1细胞、Th17细胞、Th1/B10比例和Th17/B10比例增多,而Treg细胞、B10细胞和IL-10^(+)B细胞减少(P<0.05)。C组小鼠B10细胞数量与Th1细胞和Th17细胞数量呈负相关(r=-0.394.-0.472,P<0.05),而与Treg细胞和IL-10^(+)B细胞数量呈正相关(r=0.459、0.786,P<0.05)。结论NOD小鼠存在Breg细胞与其他免疫细胞的免疫失衡,参与1型糖尿病的发生。 Objective To explore the relationship of regulatory B cells,(Breg)and other immune cells in non-obese diabetic(NOD)mice.Methods Forty-five NOD mice were equally divided into three groups of A(4 weeks),B(8 weeks)and C(type 1 diabetes).The histology of mouse pancreas was observed by HE staining.Flow cytometry was used to detect the quantities of Treg(CD4^(+)CD25^(+)Foxp3^(+)),Th1(CD4^(+)IFN-γ^(+)),Th17(CD4^(+)IL-17^(+)),B10(CD19^(+)CD5^(+)CD1d^(hi)B)and IL-10^(+)B(CD19^(+)IL-10^(+))cells in three groups.Results Compared to tgroup A,the quantities of Th1,Th17,B10 and IL-10^(+)B cells were more in group B(P<0.05).Compared to group A,the quantity of Treg was less and the quantities of Th1,Th17,B10 and IL-10^(+)B cells and the ratio of Thl to B10 cells and that of Th17 to B10 cells were higher in group C(P<0.05).Compared to group B,the quantities of Th1 and Th17 cells and the ratio of Th1 to B10 cells and that of Th17 to B10 cells were higher,while the quantities of Treg,B10 and IL-10^(+)B cells were lower in group C(P<0.05).The quantity of B10 cells was negatively correlated to that of Th1 and Th17 cells(r=-0.394 and-0.472,P<0.05),and positively correlated to that of Treg and IL-10^(+)B cells in group C(r=0.459 and 0.786,P<0.05).Conclusion Immune imbalance between Breg and other immune cells exists in NOD mice,which participates the occurrence of type 1 diabetes.
