摘要
AUX1/LAX家族是一类生长素输入载体,在植物生长发育和形态建成过程中起着重要的调控作用。利用同源比对的方法从毛竹基因组中鉴定出8个PheAUX1/LAX候选基因,进行系统进化树、基因结构、基序分析。结果表明,PheAUX1/LAX候选基因分为ClassⅠ和ClassⅡ2个亚组,其中classⅠ的候选基因包含6~7个外显子,而ClassⅡ的候选基因包含8~10个外显子。不同组织器官转录组数据分析表明多数ClassⅠ候选成员在毛竹冬笋中高丰度表达,而ClassⅡ中的PH02Gene38722.t在跳鞭中高丰度表达。通过qRT-PCR对AUX1/LAX候选基因在不同生长发育时期笋中的表达量分析表明,有6个候选基因与对照相比呈现出不同程度的上调表达,表明PheAUX1/LAX候选基因广泛参与毛竹笋的生长发育。本研究将为进一步研究毛竹AUX1/LAX基因的功能和调控机制奠定理论基础。
AUX1/LAX family members are the major auxin influx carriers,which are widely participated in plant growth and development.However,little information is available about the AUX1/LAX family in moso bamboo(Phyllostachys edulis).In this study,a total of 8 AUX1/LAX genes were identified in Moso bamboo genome.According to gene structure,conserved motif and phylogenetic relationships,the PheAUX1/LAXs were classified into two classes.The expression patterns of AUX1/LAXs based on transcriptome sequencing data indicated that four AUX1/LAX genes belonged to classⅠexhibited high expression levels in winter bamboo shoot.The PH02Gene38722.t1-a classⅡmember showed high expression level in outward-rhizome.Expression analysis based on qRT-PCR indicated that six AUX1/LAXs showed high accumulation level during moso bamboo shoot growth,indicating the significant importance of AUX1/LAXs during moso bamboo shoot growth.Our study can provide a theoretical foundation and some experimental evidence for further functional verification of the PheAUX1/LAX.
