电针联合埋线治疗对功能性消化不良大鼠NLRP3水平及肠道运动功能的影响 被引量:3

Effect of Electro-Acupuncture Combined with Catgut Embedding on NLRP3 and Intestinal Motility in Rats with FD

摘要 目的:探究电针联合埋线治疗对功能性消化不良(FD)大鼠Nod样受体蛋白3(NLRP3)水平及肠道运动功能的影响。方法:将SD大鼠随机分为空白对照组、模型组、电针组、埋线治疗组以及电针联合埋线治疗组,每组各10只。除空白对照组外,其余各组大鼠均采用高脂饲料喂养+番泻叶煎剂灌胃+束缚+游泳的方法,复制FD模型,并按分组接受不同的干预处理,观察一般行为变化,测定胃排空率以及小肠推进率,并对小肠组织进行组织学观察,免疫组化和蛋白免疫印迹法检测NLRP3在小肠组织中的表达。结果:与空白对照组比较,模型组大鼠出现饮食及活动少、被束缚时反应少、呼叫弱、毛发枯乱无光泽和大便稀溏不成形的现象,肠组织出现充血水肿及淋巴细胞-浆细胞浸润,可见大量NLRP3蛋白阳性细胞,一般行为评分、胃排空率以及小肠推进率均明显下降,差异具有统计学意义(P<0.05),NLRP3蛋白表达水平显著升高,差异具有统计学意义(P<0.05)。与模型组相比,各干预组肠组织充血水肿及淋巴细胞-浆细胞浸润情况均有不同程度的缓解,NLRP3蛋白阳性细胞均有所减少,一般行为评分、胃排空率以及小肠推进率均明显升高,差异具有统计学意义(P<0.05),NLRP3蛋白表达水平显著降低,差异具有统计学意义(P<0.05);电针联合埋线治疗组效果最为明显。结论:电针联合埋线治疗能显著提高功能性消化不良大鼠肠道运动功能,可能与其抑制NLRP3蛋白水平有关。 Objective:To explore the effect of electro-acupuncture combined with catgut embedding on the level of Nod like receptor protein 3(NLRP3)and intestinal motility in rats with functional dyspepsia(FD).Methods:SD rats were randomly divided into the blank control group,the model group,the electro-acupuncture group,the catgut embedding group and the combination group(electro-acupuncture combined with catgut embedding),with 10 rats in each group.FD model was established by the combination of high-fat diet,senna decoction,binding immobility and forced swimming.After different interventions,the general behavior changes and histology of small intestine tissues were observed,gastric empty rate and small intestine propulsive rate were calculated and the expression of NLRP3 in small intestine tissues was detected by immunohistochemistry and western blot.Results:Compared to those in the blank control group,the rats in the model group showed the manifestations of less diet and activity,less response when restrained,weak squeak,messy and dull hair and loose stool,the intestinal tissues showed hyperemia and edema and the infiltration of lymphocytes and plasma cells;a large number of NLRP3 positive cells were observed;the general behavior scores,the gastric emptying rate and the small intestine propulsive rate were all significantly decreased(P<0.05);the protein expression of NLRP3 was significantly increased(P<0.05).Compared to those in the model group,the hyperemia and edema of intestinal tissues and the infiltration of lymphocytes and plasma cells were all alleviated to various degrees;the number of NLRP3 protein positive cells was decreased;the general behavior scores,the gastric emptying rate and the small intestine propulsive rate were all increased(P<0.05),the protein expression of NLRP3 was decreased in all treatment groups(P<0.05);among which the combination group had the most obvious effects.Conclusion:Electro-acupuncture combined with catgut embedding can significantly improve the intestinal motility of the rats with FD,which may be related to the inhibition of NLRP3 protein.
