香茅醛对心肌缺血/再灌注损伤大鼠的抗氧化保护作用

摘要 目的研究香茅醛对大鼠心肌缺血/再灌注(MI/R)损伤的干预作用与机制。方法可逆性冠脉左前降支结扎缺血30 min,再灌注120 min复制MI/R模型,将SD大鼠随机分为假手术组、模型组及香茅醛低、中、高(50、100、150 mg·kg^(-1))剂量组。利用生物信号采集处理系统实时监测大鼠左心室血流动力学的变化,评估心脏收缩及舒张功能;检测血清中乳酸脱氢酶(LDH)、肌酸激酶同工酶(CK-MB)水平;检测心肌组织超氧化物歧化酶(SOD)、丙二醛(MDA)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)水平以及Nrf2、HO-1的蛋白水平。结果与模型组比较,香茅醛100、150 mg·kg^(-1)组可使LVSP和+dP/dt_(max)明显升高,LVEDP和-dP/dt_(max)降低(P<0.05),并显著降低血清中LDH、CK-MB水平及心肌组织MDA含量,同时升高心肌组织SOD、CAT和GSH-Px水平(P<0.05),提高Nrf2、HO-1的蛋白表达(P<0.05)。结论香茅醛对大鼠MI/R损伤具有抗氧化保护作用,有望成为心血管急重症治疗中预防并发症的新型手段。 Objective To determine the protection of citronellal on myocardial ischemia/reperfusion(MI/R)injury in rats.Methods An in vivo model of 30 min myocardial ischemia 30 min followed by 120 min reperfusion was made by reversibly ligating the coronary left descending artery.The SD rats were randomly divided into a sham group,an MI/R model group,and 3 citronellal(50,100,and 150 mg·kg^(-1))groups.The biological signal acquisition and processing system was used to monitor the changes in the left ventricular hemodynamics in rats,and to evaluate the cardiac systolic and diastolic function.Meanwhile,the levels of lactate dehydrogenase(LDH),creatine kinase-MB(CK-MB)in the serum,as well as superoxide dismutase(SOD),malondialdehyde(MDA),catalase(CAT)and glutathione peroxidase(GSH-Px)in the myocardial tissue were determined.The expression of Nrf2,and HO-1 protein in the myocardial tissue was detected by Western blot.Results Compared with the model group,LVSP and+dP/dt_(max) were increased,LVEDP and-dP/dt_(max) were reduced,the levels of LDH,CK-MB in the serum and MDA in the myocardial tissue were decreased,while the levels of SOD,CAT and GSH-Px in the tissue and the expression levels of Nrf2 and HO-1 protein were increased in the citronellal groups at 100,and 150 mg·kg^(-1)(P<0.05).Conclusion Citronellal has antioxidant protection effect against MI/R injury in rats,and is expected to be a new way to prevent complications in acute and severe cardiovascular diseases.
