谷子bHLH转录因子家族基因鉴定及生物信息学分析 被引量:3

摘要 碱性螺旋-环-螺旋(bHLH)转录因子家族是动植物及微生物中最大的转录家族之一,参与植物生长发育、代谢调控以及应答逆境胁迫。本研究对谷子进行全基因组搜寻,获得bHLH基因家族并研究其理化性质、系统发育、染色体分布、保守结构域以及表达模式。结果表明,从谷子转录组数据库中共鉴定出151个bHLH基因(命名为SibHLH1~SibHLH151),不均匀地分布在9条染色体上,谷子bHLHs蛋白理化性质差异较大,68.9%的bHLH蛋白呈弱酸性。谷子bHLHs蛋白具有两个保守的结构域并含有典型的H 5-E 9-R 13序列。进化树分析显示,151个bHLHs家族蛋白分为18个亚家族,谷子bHLH转录因子与水稻同源性更高,具有更近的亲缘关系。表达分析表明,谷子所有bHLHs转录因子为组成型表达,部分基因受光、温、激素等逆境因子诱导表达。 The basic helik-loop-helix(bHLH) transcription factor family is one of the largest transcription families in plants, animals and microorganisms, and is involved in plant growth and development, metabolic regulation and response to stress.In this study, a genome-wide search was conducted to obtain the bHLH gene family of millet and the physicochemical properties, phylogeny, chromosomal distribution, conserved domain and expression pattern were studied.The results showed that a total of 151 bHLH genes(named SiBHLH1-SiBHLH151 were identified from the transcriptional database of millet, which were distributed inevenly on 9 chromosomes.The physicochemical properties of bHLHs protein in millet were significantly different, and 68.9% bHLH protein was weakly acidic.Millet bHLHs protein had two conserved domains containing typical H 5-E 9-R 13 sequence.Phylogenetic tree analysis showed that 151 bHLHs proteins could be divided into 18 subfamilies, and millet bHLH transcription factors had higher homology and closer genetic relationship with rice.Further, the expression analysis showed that all bHLHs transcription factors in millet were constitutively expressed, and some genes were induced by stress factors such as light, temperature and hormone.
