β-榄香烯对前列腺癌PC-3细胞生物学行为的影响及可能机制

Effects and its possible mechanism ofβ-elemene on biological behavior of prostate cancer PC-3 cells

摘要 目的探讨β-榄香烯对前列腺癌PC-3细胞增殖、凋亡、侵袭和迁移的影响及可能机制。方法体外培养PC-3细胞,分别以25、50、100μg/mlβ-榄香烯处理后,MTT实验与平板克隆实验检测PC-3细胞增殖能力;Transwell实验检测PC-3细胞迁移和侵袭能力;流式细胞术检测PC-3细胞的凋亡率;Western blot检测PC-3细胞哺乳动物雷帕霉素靶蛋白(mTOR)、磷酸化哺乳动物雷帕霉素靶蛋白(p-mTOR)、丝氨酸/苏氨酸激酶(Akt)和磷酸化丝氨酸/苏氨酸激酶(p-Akt)的蛋白表达。结果不同浓度β-榄香烯处理后,PC-3细胞的增殖能力、迁移和侵袭能力降低,凋亡率升高(P<0.01)。不同浓度β-榄香烯处理后,mTOR与Akt的蛋白表达并无明显变化,但是p-mTOR和p-Akt的表达明显降低(P<0.01)。结论β-榄香烯抑制人前列腺癌PC-3细胞增殖、侵袭及迁移,促进凋亡。 Objective To investigate the effect and its possible mechanism ofβ-elemene on proliferation,apoptosis,invasion and migration of prostate cancer PC-3 cells.Methods PC-3 cells were cultured in vitro,and then treated with 25,50 and 100μg/mlβ-elemene,respectively.The proliferation of PC-3 cells was detected by MTT assay.Transwell assay was used to detect the migration and invasion of PC-3 cells;The apoptosis of PC-3 cells were detected by flow cytometry.The expressions of mammalian rapamycin target protein(mTOR),phosphorylated mammalian rapamycin target protein(p-mTOR),serine/threonine kinase(Akt)and phosphorylated serine/threonine kinase(p-Akt)in PC-3 cells were detected by Western blot.Results After treatment with different concentrations ofβ-elemene,the proliferation,migration and invasion of PC-3 cells was decreased(P<0.01).The apoptosis rate of PC-3 cells was increased significantly(P<0.01).There was no significant change in the expression of mTOR and Akt,but the expression of p-mTOR and p-Akt was decreased significantly(P<0.01).Conclusionβ-elemene inhibited the proliferation,invasion and migration of human prostate cancer PC-3 cells and promotes apoptosis.
