Formononetin promotes apoptosis in vulvar squamous cell carcinoma SW962 cells through the FAK/AKT/Bcl-2 pathway

摘要 目的探讨刺芒柄花素(FMN)对外阴鳞癌SW962细胞凋亡的影响及其相关机制。方法通过CCK8方法和流式细胞仪检测FMN对SW962细胞增殖和凋亡的影响;Westernblot方法检测FMN对细胞凋亡和FAK通路相关蛋白表达的影响;通过EGF激活FAK通路,检测FMN对激活后FAK通路和凋亡相关蛋白表达的影响。结果FMN呈浓度依赖性抑制SW962细胞增殖,促进SW962细胞凋亡;FMN上调Bax、Cleaved-caspase3表达,下调pFAK、p-AKT、Bcl-2表达。EGF处理后p-FAK、p-AKT、Bcl-2表达上调,Bax、Cleaved-caspase3表达下调,FMN可逆转EGF的作用。结论刺芒柄花素通过FAK/AKT/Bcl-2通路促进外阴鳞癌SW962细胞凋亡。 Objective To investigate formononetin effect on apoptosis in vulvar squamous cell carcinoma SW962 cells. Methods Formononetin effect on the proliferation and apoptosis of SW962 cells was detected by CCK8 and flow cytometry methods. The expression of apoptosis related proteins and FAK pathway proteins was examed through western-blot method. The same as protein examination after EGF treatment. Results Formononetin inhibited SW962 cells proliferation and promoted apoptosis in a concentration-dependent manner. Formononetin upregulated the expression of Bax and Cleaved-caspase3 proteins and downregulated the expression of p-FAK, p-AKT and Bcl-2 proteins. Bax and Cleavedcaspase3 protein expression was decreased and p-FAK, p-AKT and Bcl-2 protein expression was increased after EGF treatment. Formononetin reversed the effect of EGF. Conclusion Formononetin promotes apoptosis of vulvar squamous cell carcinoma SW962 cells through the FAK/AKT/Bcl-2 pathway.
