摘要
目的探讨负载姜黄素的羧甲基壳聚糖水凝胶对大鼠烫伤创面的修复作用。方法 24只SD大鼠随机分为模型对照组、姜黄素组与姜黄素-羧甲基壳聚糖水凝胶组,每组8只。HE染色和Masson染色观察组织病理改变;Western blot检测创面组织IL-1β、IL-6和IL-10表达变化。结果姜黄素与姜黄素-羧甲基壳聚糖水凝胶治疗后的创面愈合率显著提高(P<0.05);姜黄素-羧甲基壳聚糖水凝胶优于姜黄素单独治疗组。姜黄素-羧甲基壳聚糖水凝胶治疗后,创面组织上皮化明显,炎性细胞及介质浸润减少,血管新生增加,胶原纤维含量增多且排列整齐;Western blot检测结果显示,姜黄素-羧甲基壳聚糖水凝胶治疗组创面组织中促炎细胞因子IL-1β、IL-6表达水平较模型组显著降低;抑制炎症和免疫的因子IL-10表达水平较模型组显著升高(P<0.05)。结论姜黄素-羧甲基壳聚糖水凝胶可能通过调控炎症反应,从而促进血管和胶原生成,加速创面愈合。
Objective To investigate the effect and mechanism of curcumin loaded carboxymethyl chitosan hydrogel on the repair of scald wound in rats. Methods Twenty-four SD rats were randomly divided into three groups(the model group;the curcumin group;and the curcumin loaded carboxymethyl chitosan hydrogel group). 8 days after administration, tissue samples were collected from the wound, and tissue changes were observed by he staining and masson staining. western blot were used to detected the changes of associated protein. Results The wound healing rate after treatment with curcumin and curcumin-carboxymethyl chitosan hydrogel was significantly improved(P<0.05). Curcumincarboxymethyl chitosan hydrogel was superior to curcumin alone treatment. After treatment with curcumin-carboxymethyl chitosan hydrogel, the wound tissue had obvious epithelialization, the infiltration of inflammatory cells and mediators was reduced, angiogenesis was increased, and the content of collagen fibers was increased and arranged neatly. Furthermore, the expressions of IL-1β, IL-6 and IL-10 were up-regulated(P<0.05). Conclusion Curcumin-carboxymethyl chitosan hydrogel may promote the generation of blood vessels and collagen and accelerate wound healing by regulating the inflammation signaling pathway.
