桔梗多糖对S180荷瘤小鼠肿瘤生长及免疫功能的影响 被引量:5

The effects of Radix platycodonis polysaccharide on tumor growth and immune function in S180 tumor-bearing mice

摘要 为探讨桔梗多糖对S180荷瘤小鼠肿瘤生长及免疫功能的影响,于小鼠右腋皮下注射S180细胞腹水悬液构建荷瘤小鼠模型,随机分为模型对照组、环磷酰胺(0.02 g/kg)组及桔梗多糖低(0.2 g/kg)、高(0.4 g/kg)剂量组,每组10只;另取10只健康C57BL/6小鼠作为空白对照组。环磷酰胺组腹腔注射给药,桔梗多糖低、高剂量组则灌胃给药,1次/d,连续治疗14 d。通过测定瘤质量、抑瘤率、胸腺指数、脾脏指数、淋巴细胞增殖刺激指数、NK细胞活度、血清细胞因子水平及瘤组织蛋白表达水平等指标,评价桔梗多糖对S180荷瘤小鼠肿瘤生长及免疫功能的影响。结果显示,与模型对照组比较,桔梗多糖低、高剂量组的瘤质量显著降低(均P<0.05),抑瘤率显著增加(均P<0.05);与模型对照组比较,桔梗多糖低、高剂量组小鼠胸腺指数、脾脏指数、淋巴细胞增殖刺激指数、NK细胞活度、IL-2、IFN-γ水平显著增加(均P<0.05),而TLR4、髓样分化因子88(myeloid differentiation factor 88,MyD88)、NF-κB蛋白表达水平显著降低(均P<0.05)。由此,桔梗多糖可明显抑制S180荷瘤小鼠肿瘤的生长,该作用与改善机体抗肿瘤免疫功能有关。 To investigate the effects of Radix platycodonis polysaccharide(RPP) on tumor growth and immune function of S180 tumor-bearing mice, a tumor-bearing mouse model was established by subcutaneously injecting S180 cell ascites suspension in the right axilla of C57 BL/6 mice. The model mice were randomly divided into model control group, cyclophosphamide(0.02 g/kg) group, RPP low(0.2 g/kg), and high(0.4 g/kg) dose group, with 10 mice in each group;Another 10 healthy mice were treated as the blank control group. Cyclophosphamide was given by once a day intraperitoneal injection, and RPP was daily given by gavage, all the treatment persisted for 14 d. The effects of RPP on tumor growth and immune function in S180 tumor-bearing mice were evaluated by measuring multiple relevant indexes including tumor mass, tumor inhibition rate, thymus index, spleen index, lymphocyte proliferation stimulating index, NK cell activity, serum cytokine levels, and tumor tissue protein expression levels. The results showed that compared with the model control group, the tumor mass of both the low-dose and high-dose RPP groups was significantly decreased(both P<0.05) and the tumor inhibition rate was significantly increased(both P<0.05);Furthermore, thymus index, spleen index, lymphocyte proliferation stimulation index, NK cell activity, IL-2 and IFN-γ levels in the low dose and high dose RPP groups were significantly increased(all P<0.05), whereas the levels of TLR4, myeloid differentiation factor 88(MyD88) and NF-κB protein were significantly decreased(all P<0.05). In conclusion, RPP can inhibit tumor growth in S180 tumor-bearing mice, which is related to improving the mice anti-tumor immune function.
