漏斗多孔菌液体菌种培养基及其栽培条件 被引量:6

Liquid culture medium and cultivation conditions of Polyporus arcularius

摘要 为实现漏斗多孔菌资源化利用,设计单因素试验,以菌丝生物量、菌球密度和菌球直径为指标,获得漏斗多孔菌Polyporus arcularius液体菌种培养基配方为马铃薯(去皮)200g、玉米粉20.0g、蛋白胨5.0g、KH_(2)PO_(4)3.0g、K_(2)HPO_(4)1.0g,MgSO_(4)·7H_(2)O 1.5g,初始pH 5.0,并优化培养条件为装液量250mL/500mL,接种量2%,培养温度32℃,转速170r/min,种龄6d;依上述条件制备液体菌种,以固体菌种为对照,进行出菇试验,调查农艺性状,试验结果表明液体菌种接种后,菌袋满袋时间提前7d,子实体原基产生时间提前8d,头潮菇鲜重提高22.5%;配方试验结果表明,以细木屑为主料的配方产量最高,头潮菇鲜重为270.6g/袋。 A single factor test was designed to determine the mycelial biomass,mycelial spheroid density and diameter of liquid culture of Polyporus arcularius.The formula of liquid medium was 200g potato,20g corn meal,5g peptone,3.0g KH_(2)PO_(4),1.0g K_(2)HPO_(4),and MgSO_(4)·7H_(2)O 1.5g,and the initial pH was adjusted to 5.0.Optimized culture conditions were:liquid volume of 250mL/500mL,inoculation volume of 2%,culture temperature of 32℃,speed of rotation 170r/min,inoculum aged 6d.According to the above conditions,the liquid culture was prepared.The solid culture was used as the control,and the cultivation experiment was carried out to investigate the agronomic characters.The results showed that using the liquid culture as inoculum,the fruiting stage could be shifted to earlier date,and the mycelial sackful colonization time was 7d earlier than usual.The formation of primordia was 8d earlier than usual,and the fresh weight of first harvest fruiting bodies increased by 22.5%.Using fine sawdust as main substrate could harvest the highest yield,and the harvest fresh weight of fruiting bodies could reach 270.6g/bag.
