Optimizing the Extraction Process of Qingre Chushi Zhitong Liquid Based on AHP-EntropyWeight Method Combined with Central Composite Design-response Surface Test

摘要 目的:优选清热除湿止痛液的提取工艺。方法:在单因素考察的基础上,以信息熵理论结合星点设计-效应面法,对浸泡时间、提取时间、加水量及提取次数进行考察,以绿原酸、龙胆苦苷、络石苷的含量及干膏得率为评价指标,采用信息熵理论对各评价指标赋予权重系数,并计算综合评分。通过Design-Expert 8.0.6.1软件分析各因素间的交互作用,得出优化的水提取工艺,并对该工艺进行验证实验。结果:依据信息熵理论,将绿原酸、龙胆苦苷、络石苷及干膏得率的权重系数分别确定为0.16、0.76、0.05、0.03;交互作用分析结果显示,浸泡时间对综合评分影响较大。清热除湿止痛液的最佳提取工艺确定为浸泡53 min,加9倍量水,提取3次,每次105 min;验证结果显示,各指标成分含量及综合评分RSD均小于3%。结论:优化的水提取工艺稳定、可行,可为清热除湿止痛液的进一步开发利用提供参考。 Objective:To optimize the extraction process of Qingre Chushi Zhitong Liquid.Methods:On the basis of single-factor tests,information entropy theory combined with central composite design-response surface method was employed to optimize the soaking time,material-to-liquid ratio,extraction frequency,and extraction time.The content of chlorogenic acid,gentiopicroside,and tracheloside and the yield of dry paste were taken as the indexes for evaluation.The information entropy theory was employed to assign the weight coefficient of each index,and the comprehensive score was calculated.Design-Expert 8.0.6.1 was used to analyze the interaction between factors,and the optimized water extraction process was obtained,which was then verified.Results:According to the theory of information entropy,the weight coefficients of the content of chlorogenic acid,gentiopicroside,and tracheloside and the yield of dry paste were 0.16,0.76,0.05,and 0.03,respectively.Soaking time significantly affected the comprehensive score.The optimum extraction conditions:soaking for 53 min,material-liquid ratio of 1∶9,and extraction for 3 times(105 min for each time).Under these conditions,the relative standard deviations(RSDs)of the content of each index component and the comprehensive score were less than 3%.Conclusion:The optimized water extraction process is stable and feasible,and can provide a basis for the further development and utilization of Qingre Chushi Zhitong Liquid.
