摘要
目的从基因水平探讨多环芳烃(PAHs)暴露对2型糖尿病(T2DM)可能的生物信息影响因素。方法通过比较毒理基因组学数据库(CTD)中与GEO数据库中获得的PAHs影响T2DM的相关基因及化学物,对PAHs影响T2DM的关键基因进行富集分析,获取共同基因/蛋白可能参与的进程或信号通路。结果PAHs影响T2DM的相关基因共755个,200个关键基因/蛋白富集分析结果提示,关键基因/蛋白参与呼吸电子传递链、ESR介导的信号、调节胞外分泌、细胞对有机环化合物的反应、磷蛋白磷酸酶活性的正调控、对成纤维细胞生长因子的反应、自噬和蛋白质分解过程的调节等多条信号通路。结论PAHs可能在基因水平通过多通路因子影响T2DM的发生发展。
Objective To investigate preliminary bioinformatics study on the effects of polycyclic aromatic hydrocarbons(PAHs)exposure on genes of type 2 diabetes mellitus(T2 DM).Methods The interacted genes and chemicals of PAHs exposure affecting T2 DM were obtained from the comparative toxicogenomics database(CTD)and gene expression omnibus(GEO)Datasets.Enrich analysis were performed for the key genes of PAHs exposure affecting T2 DM,and further possible biological processes and pathways were obtained from common genes/proteins.Results A total of 755 interacted genes of PAHs exposure affecting T2 DM were identified,and 200 key genes were mainly involved in respiratory electron transport,ESR-mediated signaling,regulated exocytosis,cellular response to organic cyclic compound,positive regulation of phosphoprotein phosphatase activity,response to fibroblast growth factor,autophagy,regulation of protein catabolic process.Conclusion PAHs exposure may affect T2 DM development through multiple pathways at gene levels.
