摘要
目的研究香烟烟雾与慢性应激联合作用对大鼠生殖功能的影响。方法健康清洁级SD大鼠40只,雌雄各半,随机分为对照组、香烟烟雾组、慢性应激组和联合作用组,每组10只。对照组不采取任何处理;香烟烟雾组大鼠进行呼吸道静式染毒,每次10支香烟,1次/d,每次30 min;慢性应激组从5种慢性应激方案中每日随机给予一种慢性应激,每种慢性应激方式不连续应用;联合作用组每日在香烟烟雾染毒(处理同香烟烟雾组)基础上进行慢性应激处理(处理同慢性应激组);12周后,处死大鼠,计算卵巢、睾丸脏器系数;观察大鼠卵巢、睾丸组织病理学变化;测定血清卵泡刺激素(FSH)、促性腺激素释放激素(GnRH)、胰岛素样生长因子(IGF-1)、黄体生成素(LH)和雌二醇(E2)含量。结果与对照组比较,慢性应激及联合作用组大鼠卵巢脏器系数降低,差异有统计学意义(P<0.05);香烟烟雾、慢性应激及联合作用大鼠卵巢及睾丸组织有不同程度淤血及代偿性增生,卵母细胞核萎缩,睾丸组织中生殖细胞核固缩;香烟烟雾组血清GnRH、IGF-1、LH和E2含量降低,慢性应激组血清FSH、IGF-1、LH和E2含量降低,联合作用组大鼠血清FSH、IGF-1和LH含量降低,差异均有统计学意义(P<0.05)。结论香烟烟雾与慢性应激单独及联合作用会引起大鼠的生殖功能损伤。
Objective To study the combined effect of cigarette smoke and chronic stress on the reproductive function of rats. Methods Forty healthy SD rats, 50% males, were randomly divided into control group, cigarette smoke group, chronic stress group, and joint action group, with 10 rats in each group. The control group did not take any treatment;the rats in the cigarette smoke group were given static airway exposure, 10 cigarettes each time, once a day for 30 minutes each time;the chronic stress group was randomly given daily from five chronic stress programs one type of chronic stress, each chronic stress method is not applied continuously;the joint action group is treated with chronic stress after cigarette smoke exposure(the treatment is the same as the cigarette smoke group) every day(the treatment is the same as the chronic stress group). After 12 weeks, the rats were sacrificed and the ovarian and testicular organ coefficients were calculated;the histopathological changes of the ovaries and testes were observed;the serum FSH, GnRH, IGF-1, LH and E2 content were determined. Results Compared with the control group, the ovarian organ coefficient of rats in the chronic stress and combined exposure group decreased(P<0.05);cigarette smoke, chronic stress and joint action can cause different degrees of congestion, compensatory hyperplasia of rat ovarian and testicular tissues, resulting in oocyte nucleus atrophy, and reproductive cell nuclei in testicular tissues;the cigarette smoke group serum GnRH, IGF-1, LH, E2 content decreased, and the chronic stress group serum FSH, IGF-1, LH, E2 content decreased, The serum FSH, IGF-1 and LH levels of rats in the joint action group decreased(P<0.05). Conclusion Cigarette smoke and chronic stress alone or in combination can cause damage to the reproductive function of rats.
