糯玉米自然发酵液中酵母菌的分离鉴定及其特性研究

摘要 对发酵20 d糯玉米黏豆包发酵液中的酵母菌进行分离纯化,通过菌落形态观察、镜检特征和ITS rDNA序列对分离得到菌株进行鉴定,并对其生长特性和耐受性进行评价。结果表明:从糯玉米自然发酵液中分离得到的HSD F2.2001、HSD F2.2002和HSD F2.2003这3株菌均为酿酒酵母,且菌株HSD F2.2001与菌株KX951644.1具有83%同源性,菌株HSD F2.2002和HSD F2.2003与菌株KC544501.1同源性达99%。菌株HSD F2.2001和HSD F2.2002的最佳生长温度为28℃、最适p H分别为4.5和5.0、可耐受乙醇体积分数为15%、可耐受葡萄糖质量分数为55%;2种菌株最佳利用碳源均为葡萄糖,菌株HSD F2.2001最佳利用氮源为酵母粉,菌株HSD F2.2002最佳利用氮源为牛肉浸粉。因此,从糯玉米黏豆包发酵液中分离得到的酵母菌株具有良好的生长性能,可为发酵黏豆包菌株试验提供一定理论基础。 The isolation,purification and growth characteristics of yeast in 20 d fermented glutinous Nian doubao fermentation broth were investigated. The isolated strains were identified by colony morphology observation,microscopic characteristics and ITS r DNA sequence,and their growth characteristics and tolerance were evaluated. The results showed that HSD F2. 2001,HSD F2. 2002 and HSD F2. 2003 isolated from waxy corn natural fermentation broth were Saccharomyces cerevisiae,and strain HSD F2. 2001 had 83 % homology with strain KX951644. 1,and strains HSD F2. 2002 and HSD F2. 2003 had 99 %homology with strain KC544501. 1. The optimum growth temperature of strains HSD F2. 2001 and HSD F2. 2002 was 28℃,the optimum p H was 4. 5 and 5. 0,tolerable ethanol volume fraction was 15 %,and tolerable glucose mass fraction was 55 %;The best carbon source for the two strains was glucose,the best nitrogen source for strain HSD F2. 2001 was yeast powder,and the best nitrogen source for strain HSD F2. 2002 was beef powder. Therefore,the yeast strains isolated from the fermentation broth of glutinous corn sticky Nian doubao have good growth performance,which can provide a theoretical basis for the fermentation of sticky Nian doubao.
