摘要
【目的】为克服传统抗病水稻材料抗病性易于丧失的缺点。【方法】利用寄主诱导基因沉默(HIGS)技术创制水稻抗稻瘟菌的新材料。【结果】首先,选取稻瘟菌信号肽复合物中的关键亚基SEC11为靶基因,利用Gateway基因重组技术将稻瘟菌SEC11基因的干扰片段构建到pBDL03载体上获得HIGS表达载体。通过农杆菌介导的转化法,成功将SEC11基因的HIGS载体导入水稻品种日本晴中获得阳性转基因水稻植株。最后,通过水稻叶片喷雾接种试验,证明以稻瘟菌SEC11基因为靶标的HIGS转基因水稻对稻瘟菌具有显著的抗病性。【结论】本研究创制了新的抗稻瘟病水稻材料,将HIGS技术应用于抗稻瘟病分子育种提供了科学依据。
【Objective】This paper aimed to overcome the easy breakdown of conventional disease resistant plants.【Method】A new kind of transgenic rice plants was generated based on Host Induced Gene Silencing(HIGS)technology.【Results】SEC11,a key subunit of signal peptide complex in Magnaporthe oryzae,was selected for generating the HIGS expression vector by gateway technology.HIGS vector was transferred into the Japonica rice cultivar Nipponbare by agrobacterium mediated transformation to obtain transgenic rice plants.The spray assay with conidia of rice blast indicated that the HIGS transgenic rice plants were improved in rice blast resistance.【Conclusion】This study lays the foundation for molecular breading of rice blast resistant varieties with HIGS technology.
