固定化培养对纤细裸藻的生长与生理及其对废水无机氮的去除效果 被引量:1

Effects of Immobilized Culture on Growth, Physiological and Biochemical Characteristics and Inorganic Nitrogen Removal Efficiency of Euglena gracilis

摘要 【目的】本文研究固定化培养对纤细裸藻(Euglena gracilis)生理生态及净水效果,进行为期12 d对照试验。【方法】试验设置固定化组(试验组)和液体悬浮态组(对照组),分别比较2种培养模式对纤细裸藻生长(细胞密度、相对生长率)、生理生化指标[叶绿素a、叶绿素b、类胡萝卜素、总光合色素含量、超氧化物歧化酶(Superoxide Dismutase, SOD)和丙二醛(Malondialdehyde, MDA)]以及其对养殖废水中无机氮(氨氮、亚硝酸盐氮、硝酸盐氮、总氮)的去除率。【结果】试验组纤细裸藻细胞密度、光合色素含量显著高于对照组(P<0.05);两处理组SOD、MDA均表现出先升高后降低的变化趋势,试验结束时(第12天),试验组SOD显著高于对照,而试验组MDA显著低于对照(P<0.05)。试验组对废水中无机氮去除率始终较高,在试验后期差异显著(P<0.05)。【结论】固定化培养显著促进纤细裸藻的生长、光合色素的积累,提高裸藻的抗氧化能力以及对水质无机氮的去除率。 【Objective】 This paper aimed to study the effects of immobilized culture on the physiology, ecology and water purification of Euglena gracilis,and a controlled experiment was conducted for 12 days.【Method】 Immobilized group(experimental group) and liquid suspended state group(control group) were set in the experiment in order to compare two kinds of cultivation model of Euglena gracilis(cell density, relative growth rate),physiological and biochemical indexes [chlorophyll a and chlorophyll b, carotenoids, total photosynthetic pigment content, superoxide dismutase(SOD) and malondialdehyde(MDA)]as well as its removal rate of inorganic nitrogen(ammonia nitrogen, nitrite nitrogen, nitrate nitrogen, total nitrogen) in aquaculture wastewater respectively.【Result】 The cell density and photosynthetic pigment content of the experimental group were significantly higher than that of the control group(P<0.05).In addition, both treatment groups showed a trend of SOD,MDA first increasing and then decreasing.However, at the end of the test(Day 12),SOD in the experimental group was significantly higher than that in the control group, while MDA in the experimental group was obviously lower than that in the control group(P<0.05).The removal rate of inorganic nitrogen from wastewater in the immobilized group was always high, and the difference was considerable at the later stage of the experiment(P<0.05).【Conclusion】 Immobilized culture can effectively promote the growth of Euglena gracilis and the accumulation of photosynthetic pigment and also enhance the antioxidant capacity of Euglena gracilis and the removal rate of inorganic nitrogen in water quality.
