hsa_circ_0064369在食管鳞癌组织和细胞中的表达及临床意义

Expression of hsa_circ_0064369 in esophageal squamous cell carcinoma tissues and cells and its clinical significance

摘要 目的:分析hsa_circ_0064369在食管鳞癌中的表达及其临床意义。方法:采用高通量芯片技术检测6例食管鳞癌患者癌组织和对应癌旁组织中差异表达circRNA,依据差异倍数≥5、P<0.01、样本间一致性高、原始信号值相对高等这几个特征挑选出下调8.4倍的hsa_circ_0064369为研究对象。运用实时荧光定量PCR(real-time quantitative polymerase chain reaction,RT-qPCR)检测并比较43对食管鳞癌组织和对应癌旁组织及正常食管上皮细胞(NE6)和两种食管鳞癌细胞(NE3、KYSE9706)中hsa_circ_0064369的表达水平。分析hsa_circ_0064369表达水平与临床病理特征的关系;Kaplan-Meier法分析hsa_circ_0064369表达水平与生存期的关系;构建受试者工作特征(receiver operating characteristic,ROC)曲线评估hsa_circ_0064369对食管鳞癌诊断的灵敏度和特异性。结果:食管鳞癌组织中hsa_circ_0064369相对表达量(0.35±0.41)低于癌旁正常组织(1.88±1.22)(P<0.01);两种食管癌细胞中hsa_circ_0064369的表达水平(0.46±0.18,0.53±0.09)均低于正常食管上皮细胞(1.00±0.37)(P<0.05)。hsa_circ_0064369表达下调与TNM分期显著相关(P=0.022),与性别、年龄、分化程度、肿瘤最大径、肿瘤位置、淋巴结转移无统计学相关(P>0.05)。hsa_circ_0064369低表达食管鳞癌患者无进展生存期较高表达患者短;ROC曲线下面积为0.928,当截断值为0.6995时其诊断灵敏度和特异性为88.4%和90.7%。结论:hsa_circ_0064369在食管鳞癌中表达量下降,其低表达提示预后不良,是潜在的食管癌诊断和治疗的新型分子标志物。 Objective:To analyze the expression of hsa_circ_0064369 in esophageal squamous cell carcinoma and its clinical significance.Methods:The expression of circRNA was detected with circRNA microarray assay in six samples of ESCC tumor and matched adjacent tissues.Based on the following four criterion,including:Fold Change≥5,P<0.01,high consistence in samples,high original signal value,hsa_circ_0064369 was choosed as the object of study.Real-time quantitative polymerase chain reaction was uesd to detect the expression of hsa_circ_0064369 in 43 pair ESCC tissues and normal adjacent tissues,also in normal esophageal epithelial cell(NE6)and esophageal carcinoma cell lines(NE3,KYSE9706).The relationship between the expression of hsa_circ_0064369 and its clinicopathological characteristics was analysed.A receiver operating characteristic(ROC)curve was established to evaluate the sensitivity and specificity of hsa_circ_0064369 in the diagnosis of ESCC.Results:The expression of hsa_circ_0064369 in the ESCC tissues(0.35±0.41)was lower than that in corresponding adjacent normal tissues(1.88±1.22)(P<0.01).The expression level of hsa_circ_0064369 in two kinds of esophageal carcinoma cell lines(0.46±0.18,0.53±0.09)were lower than that in normal esophageal cell line(1.00±0.37)(P<0.05).hsa_circ_0064369 expression level was associated with TNM stage(P=0.022),but showed no significant differences in gender,age,differentiated degree,tumor diameter,tumor position and lymph node metastasis.The disease free survival time was significantly shorter in patients with lowly expressed hsa_circ_0064369 than that in patients with highly expressed hsa_circ_0064369.The area under the ROC curve was 0.928 and its sensitivity and specificity were 88.4%and 90.7%when the truncation value was 0.6995.Conclusion:The expression of hsa_circ_0064369 is downregulated in ESCC tissues,indicating a poor prognosis.hsa_circ_0064369 is a potential biomarker for esophageal cancer.
